The role of noncoding RNAs on cell metabolism

Abstract:

The process of degradation, metabolism and RNA quality control is important for maintaining the balance in a cell. 3´ exonuclease DIS3L2 is also involved in these processes and is associated with formation of Perlman syndrome and Wilson's tumour. DIS3L2 is involved in degradation of cytoplasmic RNA, for instance, uridylated precursors miRNA of a family let-7. Stress stimuli cause characteristic tRNA cleavage in a cell. Under the influence of a viral stress, the tRNA fragments are created which help the cells in adaptation to stress because of their function in gene silencing. The tRNA fragments which may interact with viral DNA/RNA and help the virus to spread are also created. In the thesis, I deal with the issue of DIS3L2 protein and its effect on growth of cell cultures. It further focuses on stress and its influence on a cell, particularly tRNA degradation. A stress stimulus is caused by HRV16 virus. The thesis outlines a possible involvement of DIS3L2 protein in tRNA degradation or in maturation of tRNA fragments.

Abstract:

Proces degradace, metabolismu a kvality kontroly RNA je důležitý pro udržení rovnováhy v buňce. Na těchto procesech se podílí i 3´exonukleáza DIS3L2Ľ která byla spojená se vznikem Perlmanova syndromu a Wilsonova tumoru. DIS3L2 se podílí na degradaci cytoplazmatických RNA, jako například uridylovaných prekurzorů miRNA rodiny let-7. Stresové stimuly způsobují v buňce charakteristické štěpení tRNA. Vlivem virového stresu vznikají fragmenty tRNA, které pomáhají buňkám v adaptaci na stres tím, že mají funkci genového umlčovaní, ale i fragmenty tRNA, které mohou interagovat s virovou DNA/RNA a pomáhat v jejím množení. V této bakalářské práci sa věnuji problematice proteinu DIS3L2 a jeho vlivu na růst buněčných kultur. Dále řeší problematiku stresu a jejího vlivu na buňkuĽ konkrétně degradaci tRNA. Stresový stimul je v buňce vyvolán virem HRV16. Práce nastiňuje možné zapojení proteinu DIS3L2 do degradace tRNA, případně do maturace tRNA fragmentů.
