Study the Interactions of HU Protein of \kur{Francisella tularensis} With Host Cell

Abstract:

Tato práce je součástí projektu, který má za úkol zjistit, zda DNA vazebný protein HU bakterie Francisella tularensis interaguje s hostitelskou buňkou a pokud ano, jakým způsobem by ji HU protein mohl ovlivňovat. V jednotlivých kapitolách této práce je vypracována problematika týkající se bakterie F. tularensis a jejího zařazení do taxomické nomenklatury, jsou vypsány prozatím známé faktory virulence a stručně shrnuty informace o bakteriálním genomu, ostrovu patogenity podílejícím se na virulenci a HU proteinu. Jelikož se F. tularensis řadí mezi intracelulární bakterie, je dopodrobna rozepsán proces interakce s různými typy buněk - ať nefagocytujícími nebo buňkami patřícími do monocyto-makrofágového systému. V dalších kapitolách je popsána laboratorní diagnostika F. tularensis a tularémie i jejích klinických projevů. Poslední kapitola je věnována v dnešní době hojně zkoumané problematice F. tularensis jako nástroje možného bioteroristického zneužití. Experimentální část obsahuje tyto dílčí kroky: Kultivace bakterií E. coli TOP10 a XL-1 Blue a F. tularensis subsp. holarctica FSC200. Příprava konstruktů značeného proteinu pomocí různých typů značek (GFP, histidinové (HIS) a HA kotvy) pomocí metod molekulární biologie (tzv. "overlap" PCR, PCR, restrikční štěpení, chemická transformace a elektroporace). Značený HU protein jsme prokazovaly pomocí SDS-PAGE a Western-blottingu. Studiu lokalizace proteinu v hostitelské buňce pomocí techniky fluorescenční mikroskopie předcházela izolace, kultivace a infekce myších kostně-dřeňových makrofágů (BMMs).

Abstract:

This thesis is part of a project that aims to determine if Francisella tularensis DNA binding protein HU interacts with the host cell, and if so, how the HU protein might affect it. In theoretical part of the thesis the issue concerning the bacterium F. tularensis and its inclusion in the taxonomic nomenclature is elaborated, known virulence factors are described, and information about the bacterial genome, the pathogenicity island, which is involved in Francisella virulence, and HU protein are briefly summarized. Because of F. tularensis is intracellular bacterium its pathogenesis is described in detail, including the necessary interactions with various cell types - non-phagocytic or cells belonging to the monocyte-macrophage system. In the other chapters the laboratory diagnostics of F. tularensis and tularemia with its clinical manifestations are described. The last chapter is dedicated nowadays in detail studied problematics of F. tularensis misuse as a bioreroristic tool. The experimental part contains these partial steps: Cultivation of E. coli TOP10 or XL-1 Blue and F. tularensis subsp. holarctica FSC200. Preparation of fusion protein constructs using various types of epitope labels (GFP, HIS, HA tags) by using molecular biology methods (overlap PCR, PCR, restriction digestion, chemical transformation and electroporation). Labeled protein was detected by using SDS-PAGE and imunodetection. Protein localization studies by using fluorescence microscopy within the host cells were preceded by the isolation, culture and infection of mouse bone marrow derived macrophages (BMMs).
