Detection of Biofilm on Various Contact Surfaces

Abstract:

Byla zkoumána tvorba biofilmu vybraných mikroorganismů při různých laboratorních podmínkách. Mikroorganismy byly kultivovány na mikrotitračních destičkách a na kuponech z různých materiálů (nerezová ocel, sklo, plast a keramika). Tvorba biofilmu na mikrotitračních destičkách byla detekována spektrofotometricky po obarvení krystalovou violetí. Dále byla zkoumána účinnost mechanického odstranění biofilmu z povrchu kuponů stěrem a seškrabem. Počet buněk v biofilmu byl stanoven kultivací na pevných půdách v Petriho miskách. Účinnost mechanického odstranění byla hodnocena pomocí fluorescenční mikroskopie. Mohutnost mikrobiálního nárůstu na kuponech byla také měřena pomocí luminometru SystemSURE II (měření ATP).

Abstract:

The biofilm formation was tested using various microorganisms at different laboratory conditions. Microorganisms were cultivated on microtitre dishes and on the surface of coupons of various materials (stainless steel, glass, plastic and ceramic). The biofilm formation on microtitre dishes was detected by spectrofotometer after crystal violet-staining. The efficiency of mechanical elimination of biofilm from surface by swabbing and scraping was evaluated. The cell count in biofilm was determined by cultivation on agar plates. The efficiency of mechanical elimination of biofilm was detected by fluorescent microscopy. Microbial mass presented on the surface of tested materials was estimated by measuring of ATP using luminometer SystemSURE II.
