Population genetics of Daubenton's bat in Central Europe

Abstract:

Fenomén evropské postglaciální expanze je opěrným bodem mnoha fylogeografických studií, které napomáhají vysvětlit procesy vnitrodruhové diverzity a genetické variability. Velká pozornost je také směrována k samotným glaciálním refugiím (konkrétně Pyrenejský, Apeninský a Balkánský poloostrov), ve kterých byly mnohé temperátní druhy během zalednění de facto izolovány. Z těchto refugií pak postupnou expanzí směrem na sever vznikly četné kontaktní zóny, které se často díky hybridizaci vyznačují svou alelickou bohatostí a zvýšenou diverzitou, promítající se do populační struktury mnoha druhů organismů. Modelovým organismem pro tuto práci se stal netopýr vodní (Myotis daubentonii). Tento druh je jedním z nejběžnějších a nejvíce rozšířených druhů netopýrů v Evropě a posléze celé palearktické oblasti. Studium mitochondriální a jaderné variability tohoto druhu podporuje hypotézu výskytu kontaktní zóny mezi liniemi s východní a západní distribucí, hypoteticky vzniklých izolací v Balkánském a Apeninském refugiu, v oblasti Střední Evropy. Tato práce se zabývá mapováním kontaktní zóny v oblasti České republiky a propojením informací získaných analýzou mitochondriálních a jaderných dat. Tato kontaktní zóna je dle výsledků užší pro mitochondriální než pro mikrosatelitové markery, což odpovídá předpokladu samičí filopatrie. Difuzní charakter distribuce mikrosatelitových klastrů a přítomnost směsných jaderných genotypů a cytonukleárních konfliktů naznačují intenzivní hybridizaci mezi oběma liniemi.

Abstract:

The phenomenon of European post-glacial expansion is the backbone of many phylogeographic studies, that help to explain the processes related to evolution of intraspecific diversity and genetic variability. Apreciable attention is directed to the glacial refugia (specifically Pyrenean, Apennine and Balkan peninsula), where populations of many temperate species were de facto isolated during the glacial periods. The gradual post-glacial recolonization of many species from these refugia resulted in forming numerous contact zones. As a consequence of hybridization, these zones are frequently characteristic by their increased allelic richness and diversity, which is mirrored into the population structure of these species. The model organism for this study was the Daubenton's bat (Myotis daubentonii), which is one of the most common and widespread species of bat across the whole Europe and also in the Palaearctics. Screening of mitochondrial and nuclear variability supports the hypothesis about the occurrence of a contact zone between lineages with east and west distribution in the Central Europe. These lineages hypothetically originated from populations isolated in Apennine and Balkan refugia in the Mediterranean basin during periods of glaciation. This thesis focus to the mapping of the contact zone in the Czech Republic and integrates the data obtained by mitochondrial and nuclear analysis. According to the results, these contact zone is more narrow for mitochondrial than for microsatellite markers. This fact corresponds with the assumption of female philopatry. Diffuse distribution of microsatellite clusters and presence of admixed nuclear genotypes and cytonuclear conflicts suggest intense hybridization between both lineages.
