Monitoring of stress response in plants exposed to nanoparticles

Abstract:

Použití nanočástic v posledních letech výrazně stoupá. Využití nacházejí v průmyslových odvětvích pro své jedinečné parametry jako jsou vysoká plošná hustota a malá velikost, které podmiňují odlišné chemické, elektrické a katalytické vlastnosti. Ač mají širokou škálu uplatnění, mohou mít negativní dopad na životní prostředí. Výzkumy dopadu nanočástic na rostliny a rostlinné genomy ukazují, že nanočástice mohou mít negativní dopad na rostliny a jejich biologické dráhy, včetně regulace genové exprese a buněčných funkcí. Tato bakalářská práce se zaměřuje na zkoumání účinků nanočástic na rostliny a sledování stresové odpovědi. Teoretická část této práce se zabývá mechanismy vstupu a účinku nanočástic na rostliny. Velkou pozornost věnuje přednostně odpovědi rostlin, která je vyvolána změnou genové exprese. Zahrnuty jsou také metody, kterými lze tyto změny sledovat a analyzovat tak dopad nanočástic. Praktická část se zabývá izolací RNA z rostlin. Tato metoda umožňuje získat informace o mechanismech regulace genové exprese. Izolovaná genetická informace může být následně použita například v RT-qPCR nebo za účelem sekvenace transkriptomu a vyhodnocení dopadnu nanočástic na rostliny.

Abstract:

The use of nanoparticles has been increasing significantly in recent years. They are used in industrial sectors due to their unique parameters such as high areal density and small size, which give rise to distinct chemical, electrical and catalytic properties. Although they have a wide range of applications, they can have a negative impact on the environment. Studies on the impact of nanoparticles on plants and plant genomes show that nanoparticles can have a negative impact on plants and their biological pathways, including the regulation of gene expression and cellular functions. This bachelor thesis focuses on investigating the effects of nanoparticles on plants and monitoring the stress response. The theoretical part of this thesis deals with the mechanisms of entry and effect of nanoparticles on plants. Much attention is given preferentially to the plant response, which is induced by changes in genetic expression. Also included are methods by which these changes can be monitored to analyse the impact of nanoparticles. The practical part deals with the isolation of RNA from plants. This method allows to obtain information about the mechanisms of gene expression regulation. The isolated genetic information can be subsequently used, for example, in RT-qPCR or for transcriptome sequencing to evaluate the impact of nanoparticles on plants.
