Feeding strategies of bryophagous insects

Abstract:

Bryophagie - konzumace mechorostů, představuje paralelní potravní strategii k běžnější herbivorii. Studium bryofágního hmyzu nám proto může osvětlit herbivorii z jiných úhlů pohledu. Práce je založena celkem na třech autorových článcích, z nichž jeden je zaměřen na šířku potravní niky bryofágního hmyzu, konkrétně brouků z čeledi Byrrhidae, a další dva se zabývají mikrobiotou těchto bryofágních brouků. První studie je zaměřena na hostitelskou specificitu Byrrhidae. Zmínky o meších "asociovaných" s brouky této čeledi byly dosud odvozené spíše od mechu, ve kterém byli nalezeni, než od výsledků analýzy potravy. Provedli jsme pitvy jedinců čtyř druhů čeledi Byrrhidae, abychom stanovili jejich hostitelskou specificitu a ověřili, zda mechy přítomné v jejich nejbližším okolí jsou taktéž přítomny ve střevech. Zjistili jsme, že studované druhy jsou polyfágní, větší druhová bohatost mechů se nachází v jejich střevech než v nejbližším okolí a na úrovni jedinců byly zjištěny velké rozdíly ve složení mechů v potravě a v okolí. Druhá studie se zabývá bakteriální mikrobiotou bryofágních brouků. Pomocí Illumina MiSeq jsme určili složení mikrobiomů ve střevě, tkáni zadečku a na povrchu těla dvou druhů čeledi Byrrhidae. Zjistili jsme, že střevní mikrobiomy měly největší bohatství MOTU. Dále jsme zjistili, že mikrobiomy střeva a tkáně zadečku se významně liší od složení bakteriálních společenstev z povrchu brouků, s eudominantními bakteriemi Novosphingobium, Bradyrhizobium, Ralstonia a Caulobacter, které jsou známy svými schopnostmi detoxifikace a fixace dusíku. Třetí článek rozšiřuje předchozí studii o houbovou mikrobiotu a vliv odstranění mikrobioty na fitness hostitele. Předpokládali jsme, že mikrobiota bryofágů by měla být adaptována na extrémní potravu a na stabilní prostředí mechů, vhodné pro růst potenciálních patogenů. Testovali jsme dopady odstranění bakteriální a houbové mikrobioty na líhnutí a vývoj larev u druhu Cytilus sericeus (Forster, 1771) (Coleoptera: Byrrhidae). Zjistili jsme přítomnost prospěšné fungální mikrobioty asociované s vajíčky ale nepřenášené prostřednictvím trusu, s houbami Fusarium a Penicillium jako potenicálně důležitými pro potlačení patogenů.

Abstract:

Bryophagy - consumption of bryophytes represents a parallel feeding strategy to more common herbivory. Thus, the study of bryophagous insects can help us elucidate herbivory from different points of view. The thesis is based on a total of three author's publications, of which one is focused on the diet breadth of bryophagous insects - beetles from family Byrrhidae, and two subsequent papers are focused on the microbiota of these bryophages. The first study is focused on the host specificity of Byrrhidae. The notes of moss species "associated" with Byrrhidae were so far rather related to where they were found and not to diet analysis. We dissected individuals from four Byrrhidae species to identify their host specificity and test if the moss species from their microhabitats are present in the guts. We showed that all studied species were highly polyphagous, higher moss species richness can be found in beetle guts than in their microhabitats, and at the individual level, discrepancies were present between moss consumption and occupation. The second study deals with the bacterial microbiota of bryophagous beetles. Using the Illumina MiSeq platform, we determined the composition of microbiomes in the gut, abdomen, and on the body surface of two species of Byrrhidae. We found that gut microbiomes had the highest MOTU richness. Furthermore, gut and abdomen microbiomes differed markedly from surface bacterial assemblages, with eudominant bacteria Novosphingobium, Bradyrhizobium, Ralstonia, and Caulobacter, bacteria known for their detoxification and nitrogen fixation abilities. The third paper extended the study to fungal microbiota and impact of microbiota removal on host fitness. We expected that microbiota of bryophagous insects should be adapted on bryophages extreme diet and the stable environment of moss clumps suitable for the growth of potential pathogens. We therefore tested the impact of bacterial and fungal microbiota removal on egg hatching and larval development of Cytilus sericeus (Forster, 1771) (Coleoptera: Byrrhidae). We indicated the presence of beneficial fungal microbiota associated with eggs but not transmitted through feces, with Fusarium or Penicillium as potentially important fungi for pathogen suppression.
