Transcriptomic and proteomic changes within the course of defence reaction in plants

Abstract:

Výrazný růst lidské populace a s tím spojená rostoucí spotřeba potravin a rozšiřování sídel na úkor zemědělské půdy klade stále větší nároky na efektivitu zemědělské produkce. Jednou z cest jak zvyšovat výnosy je zmírňování dopadu stresových faktorů působících na rostliny využitím jejich přirozených obranných mechanismů. V praxi to znamená například šlechtění odolných odrůd nebo potencování obranyschopnosti specifickými chemickými látkami. Pro účely této práce byl zvolen modelový organismus rajčete a jeho genotypy s rozličnou mírou rezistence vůči napadení patogeny. Na rostliny byly aplikovány elicitiny produkované oomycetami rodu Phytophthora a Pythium, biotrofní patogen Pseudooidium neolycopersici a β – aminomáselná kyselina (BABA). Byly vytipovány geny, u nichž se předpokládala účast při navození rezistence vůči dalšímu napadení. U těchto genů byla pomocí metody PCR sledována jejich bazální transkripce a transkripce po ošetření. Součástí práce byl i základní náhled na proteomické odlišností rezistentního a náchylného druhu S. lycopersicum pomocí 2D elektroforézy a MS.Bylo zjištěno, že rostliny s různou úrovní fenotypové odpovědi na stres se poměrně významně liší a to v bazální expresi genů i mírou exprese genů po aplikaci různých látek vyvolávajících obrannou odpověď. Dále bylo zjištěno, že pro navození obranné reakce je zásadní interakce elicitinu s vysokoafinitním místem na povrchu membrány, spíše než jeho schopnost vázat steroly. Studie dále ukázaly, že kyselina salicylová pravděpodobně nebude u rostlin rajčete významnou signální molekulou a mnohem významnější signální roli hraje ethylen.

Abstract:

Population growth associated with rising food consumption and reduction of arable land puts ever greater demands on agricultural efficiency. One way to increase yields is to reduce the impact of stress factors affecting plants by using their natural defense mechanisms. This means, for example, the breeding of resistant plant varieties or the support of their defense system by specific chemicals. For the purpose of this work, the tomato model organism and its genotypes with different levels of resistance were selected. Elicitins produced by oomycetes Phytophthora and Pythium, biotrophic pathogen Pseudooidium neolycopersici and β – aminobutyric acid (BABA) were applied to the plants. Genes that were predicted to be involved in inducing resistance to further infection were selected. Basal transcription of these genes and transcription after treatment were monitored by the PCR method. A part of the thesis was also a basic overview of the proteomic differences between resistant and susceptible species of tomato by 2D electrophoresis and MS.It was found that plants with various level of phenotypic stress response differ quite significantly in basal gene expression or differ in gene expression levels after application of various defense response triggering agents. Furthermore, it has been found that the interaction of elicitin with a high affinity binding site on the surface of the membrane is essential to trigger the defense reaction, rather than its ability to bind sterols. The study also showed that salicylic acid is not likely to be a significant signal molecule for tomato plants, and ethylene signaling plays a more prominent role.
