Structural studies of infection cycles of bacteriophages

Abstract:

Rozpoznání specifických receptorů na povrchu buněk, průnik obalem hostitelské buňky a přenos genomu do cytoplazmy jsou rozhodujícími počátečními kroky fágové infekce. Mechanismy rozpoznávání hostitele a doručování genomu většiny fágů však nejsou dobře známy. V této práci předkládám strukturní studie doručení genomu bakteriofága SU10 infikujícího Escherichia coli a bakteriofága phi812 infikujícího Staphylococcus aureus. Struktury virionu bakteriofága SU10 a virionu s částečně vypuštěným genome byly vyřešeny pomocí kryo-elektronové mikroskopie. Kromě toho jsme pozorovali životní cyklus SU10 pomocí kryo-elektronové tomografie a super-resolution fluorescenční mikroskopie. Kombinace těchto metod nám umožnila prokázat, že bičík fága SU10 prochází po navázání na hostitelskou buňku strukturní reorganizací. Jedná se o první zdokumentovaný případ, kdy podofág mění strukturu svého bičíku tak, aby vytvořil trysku pro doručení genomu. Vyřešili jsme strukturu bičíku a bazální destičky bakteriofága phi812 v nativním stavu a po kontrakci. Bazální destička před kontrakcí má některé z receptor-vazebných proteiny orientvané směrem k hlavičce fága. Po kontrakci se bazální destička otevře, reorganizuje se a v nové konformaci jsou všechny receptor-vazebné proteiny se přeorientované tak, že jejich receptor-vayebná místa míří směrem od fágvé hlavičky. Naše výsledky prohlubují naše znalosti o procesech fágové infekce, což může podpořit použití fágů jako prostředků fágové terapie k léčbě infekcí způsobených kmeny E. coli a S. aureus rezistentními vůči antibiotikům. Podrobné pochopení struktury bičíku a bazální destičky fágů umožňuje racionální návrh geneticky modifikovaných fágů specificky zaměřených proti konkrétním bakteriálním kmenům.

Abstract:

The recognition of specific receptors at the cell surface, penetration of the host cell envelope, and delivery of the genome to the cytoplasm are crucial initial steps of phage infection. However, the phage-host recognition and genome delivery mechanisms of most phages are not well understood. Here I present structural studies of the genome delivery of the Escherichia coli bacteriophage SU10 and Staphylococcus aureus bacteriophage phi812. The structures of the bacteriophage SU10 virion and genome release intermediate were determined using cryo-electron microscopy. Additionally, we observed the SU10 life cycle by cryo-electron tomography and super-resolution fluorescence microscopy. This combination of methods enabled us to show that the tail of phage SU10 undergoes structural reorganisation upon host cell binding. This is the first documented case of a podophage changing its tail structure to form a nozzle for genome delivery. We solved the structure of the tail and baseplate of bacteriophage phi812 in the native and post-contracted states. The pre-contraction base plate has some of its receptor binding proteins pointing towards the phage head. Upon contraction, the base plate opens, reorganises, and all the receptor binding proteins reorient to point their receptor binding sites away from the phage head. Our results deepen our understanding of phage infection processes, which may enable the use of phages as phage therapy agens to treat infections caused by antibiotic-resistant E. coli and S. aureus. Detailed understanding of phage tail and baseplate structures enables the rational design of genetically modified phages targeted against specific bacterial strains.
