Cell morphology in cerebrospinal fluid – comparision of microscopic examination and digital morphology

Abstract:

This Bachelor's thesis is focused on evaluating the morphology of cells in the cerebrospinal fluid of children's oncology patients - microscopically and using digital morphology. CellaVision DM 1200 was used to perform automatic analysis of individual samples. This analysis is a part of the hematological laboratories, where it automates time-consuming microscopy, usually performed manually. Morphological examination of oncology patients cereberospinal fluid is performed when diagnosed, but also in certain phases of treatment. The examination is performed for possible infiltration of the blasts into the central nervous system, which affects the course of treatment. The theoretical part of the work focuses on the production, function and collection of the cerebrospinal fluid with the morphological description of the present elements. The practical part of the work represents the comparison and evaluation of both methods for which cytospinous preparations of cerebrospinal fluid have been used. Cytospinous preparations have the focus on morphology of the present elements and in particular on CNS infiltration by blasts.

Abstract:

Bakalárska práca je zameraná na hodnotenie morfológie buniek v likvore u detských onkologických pacientov mikroskopicky a pomocou digitálnej morfológie. Na prevedenie automatickej analýzy jednotlivých vzoriek bol využitý prístroj CellaVision DM 1200. Tento analyzátor predstavuje súčasť hematologických laboratórií, kde automatizuje časovo náročnú mikroskopiu, obvykle vykonávanú manuálne. Morfologické vyšetrenie likvoru onkologických pacientov je vykonávané pri diagnostike ochorenia a taktiež aj v určitých fázach liečby. Vyšetrenie sa vykonáva pre možnú infiltráciu blastov do centrálneho nervového systému, čo má vplyv na priebeh a smerovanie liečby. Teoretická časť práce sa zameriava na vznik, funkciu a odber likvoru s morfologickým popisom prítomných elementov. Praktická časť práce predstavuje porovnanie a vyhodnotenie oboch metód, na ktoré boli použité cytospinové preparáty likvoru so zameraním na morfológiu buniek a to predovšetkým na infiltráciu blastov v likvore.
