Abstract:

Tato práce se věnuje rekombinantním proteinům, zejména metodám jejich purifikace. V teoretické části se zabývám expresními systémy pro přípravu rekombinantních proteinů a metodami jejich purifikace. Dále popisuji vakuolární peptidázy s podrobnějším pohledem na aminopeptidázu Ape3p a zmiňuji možnosti, kterými lze stanovit jejich aktivitu. V experimentální části se věnuji optimalizaci izolace rekombinantního proteinu Ape3p kvasinky Candida albicans, zavedení postupu jeho purifikace a zavedení metody pro stanovení jeho enzymatické aktivity.

Abstract:

This thesis deals with recombinant proteins, especially with methods of their purification. The theoretical part deals with expression systems for the preparation of recombinant proteins and methods of their purification. I also describe vacuolar peptidases with a more detailed look at the aminopeptidase Ape3p and I mention the possibilities by which their activity can be determined. In the experimental part, I discuss the optimization of the isolation of the recombinant Ape3p protein of the yeast Candida albicans, the introduction of a procedure for its purification and the introduction of a method for the determination of its enzymatic activity.
