Relationships between elements measured in seminal fluid samples by Inductively Coupled Plasma Mass Spectrometry (ICPMS) and different spermiogram parameters

Abstract:

V posledních letech dochází celosvětově ke snižování plodnosti párů. V České republice se s tímto problémem potýká přibližně 20 % párů. K poklesu plodnosti dochází jak u žen, tak i u mužů a tento pokles je způsoben mnohými faktory. Jedním z těchto faktorů mohou být i toxické prvky, které i v nízkých koncentracích negativně ovlivňují funkci reprodukčních orgánů, přídatných pohlavních žláz a ve výsledku negativně působí na spermatogenezi a parametry spermiogramu. Na rozdíl od toxických prvků jsou esenciální prvky pro plodnost nezbytné. V této práci byly měřeny koncentrace toxických (Cd, Pb, Hg, As, V, Cr, Ni) a esenciálních prvků (Zn, Se, Mn, Mo, Co, Cu, Sr) v lidské semenné plazmě metodou hmotnostní spektrometrie s indukčně vázaným plazmatem (ICP-MS). Naměřené koncentrační hladiny toxických a esenciálních prvků byly poté korelovány s parametry spermiogramu jako je pohyblivost, morfologie a koncentrace spermií. Dále byly sledovány závislosti mezi koncentrační hladinou toxických prvků a věkem, kouřením, zvýšeným BMI, délkou ejakulační abstinence a varikokélou. Bylo zjištěno, že nejvýznamněji působícím toxickým prvkem je kadmium. Další toxické prvky negativně korelovaly s objemem ejakulátu. Esenciální prvky jen slabě korelovaly s koncentrací spermií, silněji pak s pH.

Abstract:

Over the past years fertility has been decreasing globally. In the Czech Republic approximately 20 % of couples have problem with conceiving. Fertility declines in both women and men and the decline is caused by many factors. One of these factors may be toxic elements, which even in low concentrations have a negative effect on reproductive organs, male accessory glands and even on spermatogenesis and parameters of spermiogram. Unlike toxic elements essential elements are crucial for fertility. In this work the concentrations of toxic (Cd, Pb, Hg, As, V, Cr, Ni) and essential elements (Zn, Se, Mn, Mo, Co, Cu, Sr) in human seminal plasma were measured by inductively coupled plasma mass spectrometry (ICP-MS). The measured concentrations of toxic and essential elements were then correlated with spermiogram parameters such as motility, morphology and sperm concentration. Moreover, the relationships between the concentration of toxic elements and age, smoking, increased BMI, ejaculatory ab-stinence and varicocele were studied. Cadmium has been found to be the most signifi-cant toxic element. Other toxic elements negatively correlated with the volume of ejaculate. The essential elements weakly correlated with sperm concentration and strongly with pH.
