Alzheimer disease from view of molecular biology

Abstract:

Alzheimerova choroba je závažné neurodegenerativní onemocnění. Postižené osobě se postupem času zhoršují kognitivní funkce. Po určité době se už o sebe postižená osoba nedokáže sama postarat a je zcela závislá na okolí. Po sedmi až deseti letech postižený umírá. Alzheimerovu chorobu charakterizuje ukládání patologických Aβ peptidů v mozkové tkáni. Tyto peptidy způsobují nevratná poškození neuronů a jejich následnou smrt. V mozcích pacientů s Alzheimerovou chorobou lze také nalézt hyperfosforylované τ proteiny, které se shlukují do neurofirilármích klubek. Tato klubka pravděpodobně neuronům také škodí. Další faktory podílející se na Alzheimerově chorobě jsou oxidativní stres a zánět. Na vzniku a průběhu Alzheimerovy choroby se podílí velké množství genů. Ale pouze o čtyřech z těchto genů můžeme říct, že se na vzniku Alzheimerovy choroby podílí zásadním způsobem. Jsou to geny pro APP, PSEN1, PSEN2 a APOE. Další geny, které mají na toto onemocnění vliv jsou geny pro NEP, IDE, CLU, PICLAM, SORL1, ACE a RXRA. V současnosti zatím nevíme jak tuto chorobu léčit, avšak díky včasné diagnóze se daří toto onemocnění alespoň zpomalit. Pro částečnou predikci Alzheimerovy choroby je v současné době vhodná analýza 18 plazmatických signálních proteinů a analýza transkriptomu z periferní krve.

Abstract:

Alzheimer's disease is severe neurodegenerative disease. Disabled people are gradually deteriorating cognitive functions. After some time diseased person can‘t take care of himself and is totally dependent on another people. After seven to ten years, the person dies. Alzheimer's disease is characterized by aggregation of pathological Aβ peptides in brain tissue. These peptides cause irreversible damage of neurons and their consecutive death. In the brains of AD patients can also be found hyperphosphorylated τ proteins, which aggregate into neurofibrillary tangles. These tangles may be also detrimental to neurons. The other factors contributing to Alzheimer's disease are oxidative stress and inflammation. A lot of genes involve on origin and course of Alzheimer's disease. But only four of these genes contribute on emergence of Alzheimer's disease substantially. These are APP, PSEN1, PSEN2 and APOE. The other genes that effect the disease are genes for NEP, IDE, CLU, PICLAM, SORL1, ACE and RXRA. In these days we don‘t know how to treat this disease, but through to early diagnosis of this disease, we can it at least slow down it. For a partial prediction of Alzheimer's disease is in this time appropriate analysis of 18 signal plasma proteins and transcriptome analysis of peripheral blood.
