Effects of environmental contaminants on somatic testicular cells

Abstract:

ABSTRAKT (Czech) Narušení samčího reprodukčního zdraví, například zhoršená kvalita spermií, vyšší výskyt abnormalit pohlavních orgánů či rakoviny varlat, bývá v posledních desetiletích spojováno s expozicí toxickým látkám z životního prostředí, zvláště pak látkám narušujícím hormonální systém ("endocrine disruptive compounds " - EDCs). Testikulární mezibuněčná komunikace zprostředkovaná mezerovými spoji (GJIC), které jsou tvořeny konexiny, se podílí na regulaci steroidogeneze a spermatogeneze v somatických buňkách varlat a udržuje buněčnou a tkáňovou homeostázi. Hraje tedy významnou roli při udržení samčího reprodukčního zdraví. Ve studiích 1-3 jsme se proto zaměřili na testikulární GJIC jako potenciální cílové místo EDCs a s tím spojené buněčné a molekulární mechanismy. Dle našich výsledků mohou na samčí rozmnožovací soustavu skrze ovlivnění GJIC a konexinu 43 (Cx43). U polycyklických aromatických uhlovodíků (PAHs) jsme pozorovali rychlý, strukturně závislý účinek na deregulaci GJIC, který byl navíc specifický pro jednotlivé typy testikulárních buněk. Nízkomolekulární PAHs, které mají ve své struktuře „zálivy“ nebo jim podobné oblasti, rychle inhibují GJIC v Leydigových TM3 buňkách prostřednictvím relokalizace Cx43 z buněčné membrány do cytoplazmy. Oproti tomu žádný ze sledovaných PAHs neovlivnil GJIC v Sertoliho TM4 buňkách (studie 1). Naproti tomu další EDCs (alkylfenoly, PCB 153, metoxychlor, vinclozolin, triklosan, triklokarban, dichlorodiphenyltrichloroethan (DDT) a lindan) inhibovali testikulární GJIC u obou typů somatických testikulárních buněk, tedy u Leydigových i Sertoliho buněka to skrze relokalizaci Cx43 z plazmatické membrány. Toto narušení fungování GJIC bylo spojeno s pozměněnou aktivitou mitogenem aktivovaných proteinových kináz MAPK-Erk1/2 and MAPK-p38 a s poklesem produkce progesteronu (studie 2 a 3). Dvourozměrné (2D) buněčné modely jsou považovány za tradiční nástroj pro hodnocení testikulární toxicity. Nicméně buňky kultivované v jedné vrstvě na plochém podkladu neodráží trojrozměrnou (3D) fyziologii in vivo. Proto jsme charakterizovali i 3D model nezralých myších Leydigových TM3 buněk vhodný pro skrínink in vitro (sub)chronické testikulární toxicity pomocí analýzy obrazu. Naše výsledky ukázaly, že testikulární toxicita způsobená EDCs je nejen závislá na typu aplikovaného modelu (2D vs. 3D) ale také na délce expozice. Protože experimentální 3D model umožňuje delší trvání experimentu než 2D modely, vyhodnotili jsme 3D model jako vhodnější pro dlouhodobé (subchronické a chronické) studie (studie 4). Tato disertační práce přispívá k lepšímu porozumění přímého buněčného a molekulárního účinku EDCs na somatické testikulární buňky, a tedy jejich role v narušení samčího rozmnožování. Vývoj a funkce Leydigových a Sertoliho buněk mohou být těmito EDCs narušeny během prepubertálního období, což může vést k ovlivnění samčí plodnosti i vzniku rakoviny varlat v dospělosti a stáří.

Abstract:

ABSTRACT (Eng.) Exposure to environmental toxicants, especially the endocrine-disrupting chemicals (EDCs), has been associated with male reproductive health problems, including deteriorated sperm quality, increased abnormalities in male reproductive organs, infertility, and testicular cancer over the past decades. Testicular gap junctional intercellular communication (GJIC) and their building blocks, connexins, participate in the local regulation of steroidogenesis and spermatogenesis in somatic testicular cells and play a central role in cellular and tissue homeostasis and consequently in male reproductive health. Therefore, we assessed testicular GJIC as a potential target for EDCs along with the involved cellular and molecular mechanisms (Studies 1-3). Our findings confirmed that structurally different EDCs could manifest their deleterious effects in the male reproductive system via testicular GJIC and connexin 43 (Cx43). Polycyclic aromatic hydrocarbons (PAHs) showed structure-dependent and cell type-specific rapid GJIC dysregulation. The low molecular weight PAHs with a bay or bay-like region rapidly dysregulated testicular GJIC in Leydig TM3 cells by re-localization of Cx43 from the plasma membrane to cytoplasm. None of the PAHs dysregulated GJIC in Sertoli TM4 cells (Study 1). In contrast, other GJIC-inhibiting EDCs (alkylphenols, PCB153, methoxychlor, vinclozolin, triclosan, triclocarban, dichlorodiphenyltrichloroethane (DDT) and lindane) rapidly dysregulated GJIC in both Leydig and Sertoli cells via the relocation of Cx43 from the plasma membrane. This GJIC disturbance was associated with the altered activity of mitogen-activated protein kinases MAPK-Erk1/2 and MAPK-p38 and the decline of progesterone production (Study 2 and 3). Two-dimensional (2D) cell models have been considered traditional models to evaluate testicular toxicity. However, the cells cultured in monolayers on flat surfaces do not mimic the three-dimensional (3D) physiology in vivo. Therefore, we characterized a 3D model of murine prepubertal Leydig TM3 cells suitable for image-based screening of in vitro (sub)chronic testicular toxicity. Our results showed that the testicular toxicity of EDCs was dependent not only on the type of applied culture (2D vs. 3D model) but also on the length of exposure. Since the experimental design allows 3D models to be more stable and have longer life spans than 2D models, we considered 3D models as more suitable ones for long-term (subchronic and chronic) studies than 2D models (Study 4). Overall, the results of this dissertation thesis help to better understand the role of EDCs and their direct cellular and molecular actions on somatic testicular cells in male reproductive health dysfunctions. EDCs can disturb Leydig and Sertoli cells' development and functions during a prepubertal period, leading to impaired male reproductive health and even testicular cancer later on in adulthood or aging.
