Experimental study of urinary calculi formation in organic gels

Abstract:

Na základě poznatků o močových konkrementech jakožto běžném onemocnění byly v této práci studovány reakčně difuzní systémy v laboratorních podmínkách pro simulaci vzniku a stability dvou hlavních mineralogických skupin, které tvoří močové konkrementy – šťavelany a fosforečnany vápenaté. Pro experimenty v podobě klasického Liesegangova experimentu v kolonách byla jako gelové médium vybrána organická želatina. V experimentech se studoval vliv změny počátečních koncentrací vstupních látek na vznik a morfologii vzniklých krystalických sraženin. Geochemickým modelováním byly vytvořeny stabilitní diagramy jednotlivých experimentů pro určení hranic stability vysrážených krystalic-kých látek. Pomocí PXRD analýzy byly vzorky sraženin z vybraných experimentů identifiko-vány a výsledky byly porovnány s výsledky geochemických modelů. Na základě výsledků bylo potvrzeno, že nejen změna koncentrací výchozích látek má stěžejní vliv na vznik sraženiny, ale i sled látek ve dvoufázovém, reakčně difuzním gelovém experi-mentu má velmi podstatný vliv na vznik a morfologii sraženiny. V navržených experimentech se za pomoci gelového prostředí podařilo vysrážet minerály hydroxyapatit a whewellit z obou nejčastějších skupin minerálů, tvořících močové konkrementy.

Abstract:

Based on knowledge of urinary calculi as a common disease, reactive-diffusion systems were studied in laboratory conditions to simulate the formation and stability of two major minera-logical groups that form urinary stones – calcium oxalates and calcium phosphates. For experiments in the form of Liesegang experiment in columns, organic gelatin was used as the gel medium. In the experiments, the influence of the change of the initial concentrations of input substan-ces on the formation and morphology of the crystalline precipitates was studied. By geoche-mical modeling, stability diagrams of individual experiments were constructed to determine the stability boundaries of precipitated crystalline substances. Using PXRD analysis, Precipita-tes samples from selected experiments were identified and the results compared with the re-sults of geochemical models. Based on the results, it was confirmed that not only the change of the concentrations of the imput substances has a major influence on the formation of the precipitate, but also the seque-nce of substances in the reactive diffusion gel experiment has a very significant influence on the formation and morphology of the precipitate. In the experiments, the minerals hydroxyapa-tite and whewellite (from the two most common groups of minerals forming the urinary sto-nes) were able to precipitate with the help of the gel environment.
