Determination of SVCV viral expression in selected organs of cyprinid fish and their hybrids by real-time quantitative PCR.

Abstract:

Tato bakalářská práce formou literární rešerše předkládá informace o vi-rovém onemocnění kaprovitých ryb (Cyprinidae) označovaném jako jarní virémie kaprů neboli SVC (z angl. Spring Viremia of Carp) způsobovaného původcem SVCV (z anlg. Spring Viraemia of Carp Virus). Úvodní část sumarizuje poznatky o tomto onemocnění aktuálně dostup-né v odborné literatuře včetně jeho etiologie, patogeneze a rozšíření. Druhá část práce shrnuje známé metody diagnostiky a kvantifikace míry nákazy virovými onemocněními ryb, kam patří i námi využita metoda jednokrokové kvantitativní PCR v reálném čase (z angl. Quantitative Reverse Transcripti-on – Polymerase Chain Reaction, qRT-PCR). Závěrečná část rešerše srovnává míru exprese vybraných, především imunitních genů v různých orgánech hostitele ve vztahu k nemocem vyvolaným viry nebo jinými patogeny. Praktická část bakalářské práce doplňuje literární rešerši a demonstruje potenciální vliv hybridizace na míru virové nákazy. V experimentu byli pro infekci SVCV virem použiti jedinci kapra obecného (Cyprinus carpio), kara-sa stříbřitého (Carassius gibelio) a jedinci z vybraných hybridních linií těch-to druhů. K porovnání míry virové exprese v jednotlivých rodičovských a hybridních liniích byla použita metoda qRT-PCR, přičemž intenzita nákazy byla definována počtem virových kopií ve vybraných orgánech infikova-ných ryb. Výsledky naznačují, že proces hybridizace u linií F1 a zpětných hybridů má vliv na intenzitu nákazy SVCV virem a přibližuje tyto hybridy k rodičovskému fenotypu.

Abstract:

This bachelor's thesis presents information in the form of a litera-ture search on the viral disease of carp fish (Cyprinidae) known as spring viraemia of carp (SVC) caused by the causative agent of SVCV (the Spring Viraemia of Carp Virus). The introductory part summarizes the knowledge about this dis-ease currently available in the literature, including its etiology, path-ogenesis and spread. The second part of the work summarizes the known methods of diagnosis and quantification of the degree of in-fection with viral diseases of fish, which includes method of one-step quantitative PCR in real time (Polymerase Chain Reaction, qRT-PCR) that was used in this work. The final part of the research compares the rate of expression of selected, especially immune genes in different organs of the host in relation to diseases caused by viruses or other pathogens. The practical part of the bachelor's thesis complements the litera-ture search and demonstrates the potential effect of hybridization on the rate of viral infection. In the experiment, individuals of common carp (Cyprinus carpio), silver carp (Carassius gibelio) and individu-als from selected hybrid lines of these species were used for SVCV virus infection. The qRT-PCR method was used to compare the lev-el of viral expression in the individual parental and hybrid lines, the intensity of the infection being defined by the number of viral copies in selected organs of the infected fish. The results suggest that the effect of hybridization on lines F1 hybrids and back cross hybrids influence intensity of SVCV virus infection and approximate those hybrids to a parental phenotype.
