Abstract:

Tato bakalářská práce se zabývá různými experimentálními přístupy pro měření rozpustnosti a agregace proteinů v živých buňkách, které lze využít pro screening proteinových knihoven nebo pro generování rozsáhlých datových sad potřebných k tvorbě počítačových predikčních nástrojů. První část práce se věnuje pojmům rozpustnosti a agregace proteinů a také jejich počítačovým předpovědím. Druhá část je zaměřena na experimentální metody. Diskutované metody využívají filtraci, fúzi s jinými proteiny nebo krátkými peptidy, vlastní mechanismy hostitelských buněk nebo barviva specifická pro agregaci. Tyto přístupy jsou v práci detailně popsány a kriticky porovnány s uvedením jejich praktického využití.

Abstract:

This Bachelor Thesis is focused on different experimental approaches for protein solubility and aggregation measurements in living cells. These methods can be used for protein library screening or for the collection of large data sets that are essential for the development of computational prediction tools. The first part of the Thesis deals with the concepts of protein solubility and aggregation and their computational predictions. The second part is focused on the experimental methods. Reviewed methods utilize filtration, protein or small peptide fusions, innate host cell mechanisms or aggregation-specific dyes. In the Thesis, such experimental assays are described in detail, critically compared and their practical applications are discussed.
