The role of long-non-coding RNAs in animal cells and cancerogenesis

Abstract:

Nowadays, it seems that protein coding genes in humans and higher animals are just 1,5% of the genome, but more than one third of all genes is transcribed into RNA. This noncoding genes can have important functions in regulations of cell processes and their aberrant expression was described in many cancer types and other diseases. This bachelor thesis deals with long noncoding RNAs (lncRNA), which are defined as RNAs longer than 200 nucleotids. There are described general mechanisms of how lncRNAs work, examples in animal cell physiology (apoptosis, proliferation, differentiation) and summary of their importance in cancerogenesis with emphasis on hematological malignities.

Abstract:

V současnosti se zdá, že množství genů kódujících proteiny tvoří jen cca 1,5% genomu člověka a vyšších živočichů, přičemž až třetina genomu je přepisovaná do RNA. Takovéto proteiny-nekódující RNA mohou mít zásadní funkce v regulaci buněčné fyziologie a jejich aberantní exprese byla popsána u mnoha nádorových i nenádorových chorob. Bakalářská práce se zabýva skupinou dlouhých nekódující RNA (long-non-coding RNA, lncRNA), které jsou obvykle definovány délkou větší než 200 nukleotidů. Popisuje obecné mechanismy jejich působení, příklady jejich významu ve fyziologii živočišných buněk (např. apoptóza, proliferace, diferenciace) a shrnuje jejich význam pro kancerogenezi s důrazem na hematologické malignity, kde již byl jejich význam popsán.
