Non-oxime reactivators of organophosphate-inhibited cholinesterases

Abstract:

Bakalářská práce je zaměřena na alternativní cesty při léčbě intoxikace organofosfáty inhibující funkci cholinesteras v organismu. První část práce je zaměřena na cholinesterasy, na jejich funkci, strukturu a samotnou inhibici. Dále jsou popsány komerčně využívané reaktivátory a práce je následně soustředěná hlavně na reaktivátory organofosfáty inhibovaných cholinesteras, které ve své struktuře neobsahují oximovou skupinu, avšak obsahují jiné nukleofilní centrum. U jednotlivých skupin jsou uvedeny jejich klady i nedostatky a porovnání terapeutického využití. Mimo klasickou terapeutickou metodu, jakou je aplikace atropinu a oximové léčby, jsou v současnosti vyvíjeny nové sloučeniny bez oximového fragmentu, představující další možnost terapie po otravě organofosfáty. V práci jsou uvedeny jejich struktury, racionální design a popsané publikované reaktivační schopnosti. Cílem této práce je poskytnout komplexnější přehled nových léčebných postupů se zaměřením na neoximové reaktivátory a zhodnotit tak jejich potenciál aplikace v praxi.

Abstract:

The bachelor thesis is focused on alternative pathways in the treatment of organophosphate intoxication inhibiting the function of cholinesterases in the body. The first part of the thesis is focused on cholinesterases, their function, structure, and inhibition. Next, commercially used reactivators are described and the work is then mainly focused on reactivators of organophosphate-inhibited cholinesterases, which do not contain an oxime group in their structure but contain a different nucleophilic centre. The advantages and disadvantages of classical therapeutic method, such as the application of atropine and oximes, new compounds without the oxime fragment are currently being developed, representing another therapeutic option after organophosphate poisoning. Their structures, rational design and published reactivation abilities are described. The aim of this work is to provide a more comprehensive review of new therapies with a focus on non-oxime reactivators and thus to evaluate their potential for application in practice.
