Production of human glycosytransferases in yeast expression systems

Abstract:

Glykosylace je jednou z nejdůležitějších posttranslačních modifikací, která propůjčuje proteinům zcela nové vlastnosti. Enzymy zodpovědné za glykosylace se nazývají glykosyltransferasy. Jedná se o širokou skupinu enzymů, která katalyzuje postupné syntetizování oligosacharidových řetězců připojených k různým biomolekulám, nejčastěji proteinům. Proces glykosylace umožňuje proteinům vykonávat nejrůznější funkce hrající zásadní role v živých organismech. Při patologických stavech může docházet k neřízené expresi některých glykosyltransferas. To bylo v minulosti prokázáno u N-acetylglukosaminyltransferasy V (GnT-V) a Core 2 N-acetylglukosaminyltransferasy I (c2GnT-I). Ty mají zvýšenou expresi v rakovinných buňkách, což má vliv na metastazování a rozvoj rakoviny. Přípravu a studium těchto enzymů znesnadňuje fakt, že se jedná o nestabilní, často transmembránově vázané proteiny, jež se vyskytují v malých množstvích. Jako modelový enzym při jejich studiu slouží N-acetylglukosaminyltransferasa I, jejíž 3D struktura byla nedávno vyřešena. Tato práce se zabývá studiem lidských glykosyltransferas (GnT-I, GnT-V a c2GnT-I) v eukaryotním expresním systému Saccharomyces cerevisiae s upraveným typem glykosylace. Využíváme syntetické geny kódující katalytické domény. V průběhu práce byly vyzkoušeny různé techniky klonování DNA. Exprese připravených konstruktů byla prováděna v lahvích a laboratorním fermentoru. Následná desintegrace buněk byla prováděna pomocí skleněných kuliček, emulsiflex C3 homogenizátoru a fastprep homogenizátoru. Při snaze o detekci a identifikaci studovaných glykosyltransferas byly vyzkoušeny různé způsoby a podmínky exprese, rozbíjení buněk a detekce. Přesto exprese GT genů v Saccharomyces cerevisiae nebyla doposud ověřena.

Abstract:

Glycosyltransferases are a broad group of enzymes responsible for glycosylation. It is one of the most important post-translational modifications, which provides new properties to proteins. Glycosyltransferases catalyze a sequential synthesizing of oligosaccharide chains, that are attached to various biomolecules, mostly proteins. The glycosylation enables proteins to perform various essential functions in organisms. However, pathological conditions can lead to uncontrolled expression of some glycosyltranferases for example N-acetylglucosaminyltransfese V and core 2 N-acetylglucosaminyltransferase I. Increased expression of these enzymes has been detected in cancereous cells and it was shown they affect the cancer development and metastasis. Due to the fact GTs are unstable and often transmembrane bound proteins, their preparation and studying are accompanied with several difficulties. Additionally, in the cells they are presented in small quantities. N-acetylglucosaminyltransferase, whose 3D structure was recently revealed, was chosen as a model enzyme for the study of other glycosyltransferases. This work was focused on the study of human glycosyltransferases (GnT-I, GnT-V and c2GnT-I) in an eukaryotic expression system Saccharomyces cerevisiae with a modified type of glycosylation. We used synthetic genes encoding catalytic domains and we tried various techniques of cloning DNA for preparing constructs. Expression of constructs was performed in cultivation bottles and a laboratory bioreactor. The subsequent disintegration of cells was performed using glass beads, FastPrep homogenizer and emulsiflex C3 homogenizer. Various ways and conditions of expression, cell disruption and detection to were performed to detect and identify studied glycosyltransferases. Despite all efforts GT genes expression has not been confirmed.
