Interleukin-17 in pig

Abstract:

Interleukin 17 je prozánětlivý cytokin produkovaný převážně aktivovanými CD4+ a γδ+ T lymfocyty. Jeho produkce těmito buňkami je zvýšena mimo jiné i po infekci organismu Gram-negativními bakteriemi. Cílem této diplomové práce bylo zjistit, zda prasečí mononukleární buňky periferní krve (peripheral blood mononuclear cells = PBMC) odpovídají na stimulaci bakterií Salmonella enterica produkcí IL 17 a pokud ano, tak který bakteriální faktor je za tuto stimulaci zodpovědný. Dalším cílem bylo zjistit, která populace buněk je hlavním zdrojem IL 17 po dvouhodinové stimulaci těchto buněk bakterií. Proto byly prasečí PBMC dvě hodiny stimulovány bakterií Salmonella enterica sérovary Typhimurium, Derby, Enteritidis a Infantis, dále pak jejími mutanty s chybějícími ostrovy patogenity ΔSPI 1, ΔSPI 2, s chybějícím genem pro flagelin ΔfliC, s mutacemi v genu pro lipopolysacharid ΔrfaL, ΔrfaG, ΔrfaC, v genu pro hlavní podjednotku tenkých stočených fimbrií ΔCsgA, v porinu vnější membrány ΔOmpC a regulátoru, který řídí jeho expresi ΔOmpC. Dále byly buňky stimulovány solubilním flagelinem, solubilním lipopolysacharidem (LPS), bakterií Escherichia coli a tepelně inaktivovanými bakteriemi E.coli i S. enterica. Byla měřena hladina IL 17 na úrovni mRNA za využití kvantitativní real-time PCR reakce (qRT-PCR). Odpovědi buněk na různé způsoby stimulace byly porovnávány za účelem nalezení faktoru, který je zodpovědný za stimulaci buněk k produkci IL 17. Pro jeden experiment byly využity imunomagneticky roztřízené populace CD45+, CD3+, CD4+ a γδ+ buněk za účelem nalezení konkrétní populace, která IL 17 produkuje. Bylo zjištěno, že IL 17 produkují převážně γδ+ T lymfocyty, že produkce není závislá na schopnosti bakterie vstoupit do nefagocytujících buněk ani na její schopnosti přežít v infikovaných buňkách. Dále bylo zjištěno, že stimulujícím faktorem nejsou námi zkoumané PAMP (pathogen-associated molecular pattern) – flagelin, LPS a CsgA – a odpověď buněk je rovněž nezávislá na přítomnosti porinu OmpC a jeho regulátoru OmpR. Tepelně inaktivované bakterie však nestimulují PBMC k produkci IL 17, což napovídá, že hledaný faktor je buď tepelně nestabilní, nebo je závislý na přítomnosti živé bakterie.

Abstract:

Interleukin 17 is a pro-inlammatory cytokine. The main sources of this cytokine are activated CD4+ and γδ+ T cells. Expression of IL-17 is up-regulated in response to infection of organisms by Gram-negative bakteria. The aim of the study was to find the salmonella-related factor which is resonsible for IL-17 production by porcine PBMC. Another aim was to find the population of cells which is the main source of IL 17. Swine peripheral blood mononuclear cells (PBMC) were stimulated for two hours by Salmonella enteritica serovars Typhimurium, Derby, Enteritidis and Infantis, and its mutants with deletion of pathogenity islands ΔSPI 1, ΔSPI 2, with deletion of flagelin gene ΔfliC, with deletions of some parts of lipopolysacharide gene ΔrfaL, ΔrfaG, ΔrfaC, with deletions of the major subunit of thin curled fimbriae gene ΔCsgA, with deletions of porin gene ΔOmpC and its regulator gene ΔOmpR. PBMC were also stimulated by soluble flagellin, soluble lipopolysacharide (LPS), bacteria Escherichia coli and heat-inactivated bacteria E.coli and S. enterica. The level of IL-17 mRNA was measured using quantitative real-time PCR (qRT-PCR). The immunomagnetic sorted cell populations of CD45+, CD3+, CD4+ a γδ+ cells were used in another experiment with the aim to find the population which is the main source of IL 17. It was found that γδ+ T-cells were the main source of IL 17. The production of this cytokine was independent of the bacteria ability to get into non-phagocyting cells as well as of Salmonella ability to survive inside the infected cells. The stimulating factor did not belong to PAMP (pathogen-associated molecular pattern) that was tested – flagellin, LPS and CsgA. The PBMC response was also independent of the presence of porin OmpC and its regulator OmpR. Heat-inactivated bacteria did not stimulate the PBMC to production of IL-17. Therefore salmonella-related factor which is resonsible for IL-17 production seems to be heat-unstable molecule or some factor which is dependent on presence of live bacteria.
