In Vitro Testicular Toxicity Models

Abstract:

Tato bakalářská práce řeší problematiku testování látek způsobujících testikulární toxicitu. Tyto látky narušují mužský reprodukční systém, což může vést až k neplodnosti. V teoretické části práce se porovnává tradiční přístup využívající zvířata (metody in vivo) s novým přístupem zaměřeným na tzv. alternativní in vitro modely a metody, ať již validované, v procesu validace nebo teprve ve vývoji. In vitro modely umožňují studovat účinky testovaných látek na buňky či tkáně a lépe porozumět složitým procesům odehrávajícím se ve varlatech. Je třeba pamatovat na to, že in vitro model postihuje pouze část všech interakcí a procesů odehrávajících se v in vivo prostředí varlat, proto nemůže nikdy jeden in vitro model či metoda nahradit testování na zvířatech. Stěžejní rolí budou ovšem hrát v tzv. stupňovitém schématu testování spolu s metodami in silico, ex vivo a zatím i in vivo. V práci jsou uvedeny dostupné in vitro modely ke studiu testikulární toxicity a v přehledné formě jsou shrnuty jejich výhody, nevýhody a použití. Účelem experimentální části bylo podílet se na optimalizaci dvou pokročilých in vitro testikulárních modelů. Prvním byl in vitro model ko-kultivace Sertoliho TM4 a Leydigových TM3 buněk, umožňující hlouběji zkoumat parakrinní komunikaci mezi těmito důležitými typy somatických testikulárních buněk. Druhý cíl praktické části byl zaměřen na tvorbu multicelulárních sféroidů tvořených Leydigovými TM3 buňkami.

Abstract:

This bachelor thesis addresses the issue of testicular toxicity assessment testicular. Testicular toxicants cause adverse effects on male reproductive system, which can lead to infertility. In the theoretical part of this thesis, the conventional approach using animals (in vivo methods) is compared with the new approach focused on new alternative methods including those which have been already validated or those which are being in the process of validation or development. In vitro models enable to study effects of chemicals on cells or tissues, which allows understanding the complex processes in testes. However, they also have a major disadvantage: cells or tissues are removed from their natural environment, thereby eliminating the interaction and protection mechanisms otherwise available from the donor organism. Thus, even the most powerful in vitro model or method cannot replace the whole animal testing, however, their crucial role is in a tiered testing strategy together with in silico, ex vivo and yet in vivo methods. This thesis summarizes and describes available in vitro testicular toxicity models, their advantages, disadvantages and application. The main goal of the experimental part of this work was participation in the optimization of two advanced in vitro testicular models. The first goal was an in vitro co-cultivation model of Sertoli TM4 and Leydig TM3 cells allowing studying paracrine communication between these important types of somatic testicular cells. The second goal was focused on the formation of multicellular spheroids by Leydig TM3 cells.
