Abstract:

Předmětem této dizertační práce je studium biologického významu RNA editujících enzymů z rodiny ADAR se zvláštním zaměřením na jejich roli ve vrozené imunitě a fyziologii mozku. Výsledkem doktorandských studií autora jsou čtyři publikace, které tvoří rámec této práce. Jazykem práce je angličtina. První část práce poskytuje základní informace o rodině enzymů ADAR a o RNA editaci, kterou katalyzují, s přihlédnutím k tématům autorových publikací. Následující část shrnuje výsledky publikované v autorových výzkumných článcích a rozvádí diskuzi o jejich dopadech. Poslední část pak uzavírá celou práci a je následována přílohami obsahujícími plné texty autorových publikací. Z vědeckého hlediska má tato práce dvě hlavní témata. Prvním je výzkum role myších genů Adar a Trp53 při rozpoznávání nukleových kyselin a ve vrozené imunitní odpovědi s použitím buněčných kultur myších embryonálních fibroblastů. Druhým tématem je in vitro studie mutací v lidském genu ADARB1 objevených u pacientů s mikrocefalií, epilepsií a s opožděným vývojem.

Abstract:

The subject of this thesis is the study of the biological significance of the ADAR family of RNA editing enzymes with a particular focus on their roles in innate immunity and brain physiology. The author’s doctoral studies yielded four publications that form the framework of the thesis. The first part of the thesis provides basic information about the ADAR family of enzymes and the RNA editing that they catalyse with regards to the topics of the author’s publications. The next part summarises the results published in the author’s research articles and discusses their implications. The last part concludes the thesis and is followed by annexes containing full texts of the author’s publications. From a scientific standpoint, there are two main topics of the thesis. The first one is an investigation of the role of mouse genes Adar and Trp53 in nucleic acid sensing and innate immune response using cultured murine embryonic fibroblasts. The second one is an in vitro study of human ADARB1 gene mutations discovered in patients with microcephaly, epilepsy and developmental delay.
