Mechanisms of high temperature tolerance in succulent plants

Abstract:

Sukulentní rostliny se často vyskytují na stanovištích, kde jsou vystaveny nepříznivým abiotickým faktorům vnějšího prostředí. Tato práce popisuje podrobně mechanismy, kterými se vyrovnávají s vysokými teplotami. Pro přežívání vysokých teplot, které běžně rostliny netolerují, slouží několik skupin mechanismů. První skupinou jsou morfologické a anatomické adaptace. Velikost, tvar, vnitřní stavba a vysoký obsah vody v orgánech propůjčují sukulentům vysokou tepelnou kapacitu. Silná kutikula nepropustná pro vodu, která společně s nejrůznějšími formami trichomů odráží dopadající záření je dalším důležitým znakem. Asimilační cesta CAM je jedním z hlavních fyziologických mechanismů, který napomáhá sukulentům v prostředí vysokých teplot a nízké dostupnosti vody. Kromě úspornějšího hospodaření vodou sukulentům umožňuje větší termostabilitu důležitých enzymů zapojených do fixace uhlíku. Velmi důležitou roli v odolnosti k vysokým teplotám hraje u sukulentů termostabilita buněčných membrán. Díky většímu poměru nasycených mastných kyselin v membránových lipidech si dokážou membrány udržet požadovanou fluiditu a setrvat tak ve funkčním stavu i při vysoké teplotě. Poslední popisovanou skupinou mechanismů působící na buněčné úrovni a umožňující přežívání sukulentů při vysokých teplotách je zvýšená syntéza HSP (Heat Shock Proteins). Jedná se o polypeptidy s funkcí chaperonů, které napomáhají obnovit strukturu teplem poškozených proteinů. V případě jejich nevratného poškození zabraňují jejich hromadění v buňce stimulací jejich rozkladu. Zrychlené zvýšení hladiny HSP je důležitý prvek při budování vyšší termotolerance v procesu aklimatizace.

Abstract:

Succulent plants often grow in habitats with adverse abiotic environmental factors. This thesis describes in detail the mechanisms that allow succulents to survive in high temperatures successfully. Several groups of acclimatory mechanisms can help succulents to survive high temperatures, which plants do not normally tolerate. The first group are morphological and anatomical adaptations. The size, shape, internal structure and high water content of the organs confer a high heat capacity on succulents. Another essential feature is the thick, water-impermeable cuticle, which, together with various types of trichomes, reflects incident radiation. The CAM assimilation pathway is one of the main physiological mechanisms that help succulents in dry and hot environments. In addition to more efficient water use, it provides succulents with greater thermostability of essential enzymes involved in carbon fixation. The thermostability of the cell membranes of succulents plays a crucial role in their resistance to high temperatures. Due to the higher content of saturated fatty acids in membrane lipids, membranes can maintain the required fluidity and remain functional even at high temperatures. The last group of described protective mechanisms acting at the cellular level and enabling succulent survival at high temperatures is the increased synthesis of HSPs (Heat Shock Proteins). These polypeptides with chaperone function help to restore the structure of heat-damaged proteins. In case of their irreversible damage, they prevent their cell accumulation by stimulating their decomposition. The accelerated increase in HSP levels is an essential element in building higher thermotolerance in the acclimation process.
