Abstract:

Tato diplomová práce se zabývá stanovením změn aktivit některých klíčových enzymů v metabolismu thyreoidálních hormonů u laboratorních potkanů, v závislosti na různém farmakologickém ovlivnění pokusných zvířat. Cílem experimentální části bylo zjistit vliv podávání statinů (STAT), červeného palmového oleje (RPO) a n-3 polynenasycených mastných kyselin s dlouhým řetězcem (n-3 PUFA) v dietě při různých, experimentálně navozených nefyziologicky vysokých nebo nízkých sérových hladinách thyreoidálních hormonů na specifické enzymové aktivity jodothyronin dejodas D1, D2 a D3 v různých tkáních potkanů kmene Wistar, Wistar Kyoto a SHR. Zaměřili jsme se zejména na stanovení dejodas v Harderových žlázách. Monitorování thyreoidálního stavu zvířat bylo prováděno stanovením koncentrací celkových a volných frakcí thyreoidálních hormonů v krevních sérech s použitím metody RIA. Pro stanovení enzymových aktivit dejodas byly použity v laboratoři zavedené radiometrické metody. Výsledky stanovení enzymových aktivit a jejich změn v závislosti na změnách thyreoidálního stavu u kontrolních potkanů, držených na standardní dietě, odpovídají očekávaným výsledkům. Vliv podávání STAT, RPO a n-3 PUFA na sledované enzymové aktivity není příliš výrazný, ale obecně platí, že STAT, RPO a n-3 PUFA potlačují nežádoucí účinky vyvolané příliš vysokými nebo příliš nízkými hladinami thyreoidálních hormonů.

Abstract:

This thesis deals with the determination of changes in activities of several key enzymes of thyroid hormones metabolism in the rat, depending on different pharmacological treatments of experimental animals. The aim of the experimental part was to determine the influence of the administration to the rats of statins (STAT), red palm oil (RPO) and n-3 polyunsaturated fatty acids (n-3 PUFA) in diet at different, experimentally adjusted non-physiologically high or low serum levels of thyroid hormones, on specific enzyme activities of the key enzymes of thyroid hormones metabolism, the iodothyronine deiodinases D1, D2 and D3 in various tissues of Wistar, Wistar Kyoto and SHR rats. We focused on the determination of iodothyronine deiodinases activities in harderian glands. Monitoring of thyroid statuses was performed by determination of total and free fractions of thyroid hormones in samples of blood serum, using radioimmunoanalysis. Proved radiometric enzyme assays were used for the determination of iodothyronine deiodinases activities. The results of determination of enzyme activities and their changes according to changes of thyroid state in control rats, that were kept on a standard diet, corresponded with confirmed results. The effects of STAT, RPO and n-3 PUFA administration on the measured enzyme activities are not too marked, but in general, STAT, RPO and n-3 PUFA restrains undesirable effects, caused by too high or too low serum levels of thyroid hormone.
