Comparison of growth parameters of early embryos of various mouse strains.

Abstract:

Buněčný cyklus blastomer je podobný cyklu somatické buňky s tím rozdílem, že G0 fáze se u blastomer nenachází. Stejně jako cyklus somatické buňky se cyklus blastomer skládá z fází G1, S, G2 a M fáze a z kontrolních bodů, které dohlížejí na průchod buňky buněčným cyklem. Oba typy buněčných cyklů jsou regulovány cykliny a cyklin dependentními kinázami. Rozdíl existuje v délkách trvání jednotlivých fází G1, S, G2 a M fáze během průchodu buněčným cyklem časného embrya. Délka těchto fází může hrát důležitou roli během pre-implantačního vývoje a může se lišit mezi jednotlivými kmeny myší. Během časného embryonálního vývoje nedochází k interfáznímu růstu blastomery embrya a důsledkem toho dochází k zmenšování velikosti blastomer. Tato diplomová práce je zaměřena na studium rozdílů velikostí blastomer a buněčných jader v časném embryonálním vývoji u dvou běžně používaných kmenů myší (C57Bl/6 a CD1) v osmibuněčném stádiu. Pro měření objemu blastomer jsme použili rozdílné fluorescenční barvení cytoplazmy a jádra a pomocí konfokální mikroskopie jsme měřili rozdíl mezi velikostí blastomer různých kmenů. Následně jsme použitím časosběrného experimentu měřili rozdíl v čase přechodu ze čtyřbuněčného embrya do osmibuněčného stádia. Obrazová data byla analyzována pomocí softwaru IMARIS. Zjistili jsme, že u kmene C57Bl/6 měly blastomery osmibunněčného embrya vyšší poměr nukleoplazmy k cytoplazmě, vyšší cytoplazmatický objem a čtyřbuněčná embrya rychleji přecházela do osmibuněčného stadia. Výsledky naší studie naznačují, že rozdílné genetické pozadí může mít vliv na průchod buněčným cyklem.

Abstract:

The difference between somatic and blastomer cell cycle is the absence of G0 phase but remaining phases are similar in both of the cell cycle. It consists of G1, S, G2 and M phases and from control checkpoints which regulate a journey of a cell through the cell cycle. Both types are regulated by cyclins and cyclin dependent kinases, however, there is a difference in the length of G1, S, G2 and M phases in the cell cycle. Duration time of each phase may play an important role in the pre-implantation development and can vary for different mouse strains. At the early stages of the embryo development in the mammalian cell cycle, balstomeres do not grow in interphase which leads to smaller size of blastomer. This thesis predominantly focuses on to study the blastomer and nucleus size differences in the early embryo development between two commonly used mouse strains (C57Bl/6 and CD1) at an eight-cell stage. To measure the volume of blastomeres, we applied different fluorescent staining of cytoplasm and the nucleus and through using confocal microscopy we measured the difference in size of blastomeres between various mouse strains. Afterwards, through timelapse, differences in the length of time it took to change from a four-cell to an eight-cell embryo were observed. The obtained images were analysed using IMARIS software. We observed that C57Bl/6 strain had higher NC ratio, larger volume of cytoplasm and the four-cell embryos entered into eight-cell stage earlier than CD1. Findings of this study indicate that different genetic backgrounds may have an impact on development of the cell cycle.
