Population and population-genetic aspects of biological invasions and their impact on the co-evolutionary relationship between bitterling and their mussel hosts.

Abstract:

Invaze nepůvodních druhů často ohrožují původní druhy, společenstva a ekosystémy. Antropogenní faktory mohou mít výrazný dopad na současné rozšíření vnitrodruhové genetické diverzity původních i nepůvodních organismů. Přítomnost nepůvodních druhů může také ovlivňovat mezidruhové koevoluční vztahy ve společenstvech původních organismů. Interagující druhy často existují v prostorově strukturovaných populacích, ve kterých probíhá koevoluce různou rychlostí a to vytváří geografickou mozaiku adaptací a proti-adaptací. Cílem této práce bylo studovat populační a populačně-genetické aspekty biologických invazí a jejich vliv na hostitelsko-parazitické vztahy za použití behaviorálních a molekulárních přístupů. Jako studovaný systém byl použit oboustranný vztah mezi hořavkami (Rhodeus amarus) a mlži z čeledi Unionidae, který je ovlivněn recentními invazemi škeble asijské (Sinanodonta woodiana) a slávičky mnohotvárné (Dreissena polymorpha). Molekulární přístupy byly použity k odhadu genetické diverzity a struktury u původních populací hořavky duhové a zavlečených populací škeble asijské. Behaviorální metody byly použity pro studium toho, jak invaze slávičky mnohotvárné ovlivňuje mikroevoluční procesy při výběru hostitele hořavkou duhovou a jejich populační důsledky. Analýza mitochondriální DNA hořavek odhalila šest dobře podpořených linií, přičemž čtyři z nich jsou svým výskytem geograficky omezeny v egejské, kaspické a kavkazské oblasti. Tomu dobře odpovídá geografické rozšíření skupin zjištěných analýzou jaderných mikrosatelitů, která ukazuje podrobnější strukturování populací. Výrazná populační struktura dokládá důležitou roli genetického driftu při diferenciaci populací hořavek. Výsledky také ukazují přetrvání evropských hořavek ve více malých izolovaných populacích během glaciálu. Z této refugiální oblasti jen dvě linie kolonizovaly většinu kontinentální Evropy a potkaly se v západní a střední Evropě. Ukázalo se, že hranice povodí (resp. úmoří) byly překonány za pomoci člověka, což bylo pravděpodobně spojeno s chovem kapra obecného a managementem rybářských revírů. Byla zjištěna nízká mitochondriální diverzita škeble asijské u nepůvodních populací v Evropě, kde se vyskytoval jeden haplotyp, který odpovídal široce rozšířenému haplotypu v centru jejich přirozeného výskytu v Číně, odkud mohlo dojít k primární introdukci. Analýza mikrosatelitových lokusů ukázala vysoký stupeň promíchání mezi evropskými populacemi s intenzivním a stále probíhajícím genovým tokem. Populačně-genetické Bayesiánské modelování (ABC) ukázalo komplexní vztahy mezi populacemi v rámci nepůvodního areálu v Evropě. Nebyl zjištěn vliv recentní sympatrie mezi hořavkou a slávičkou na volbu hostitele a jen slabý vliv slávičky na reprodukční úspěšnost hořavky. Samice zkoumaly infikované i neinfikované velevruby ve stejné míře, ale preferovaly kladení do neinfikovaných hostitelských mlžů. Samice hořavek se úplně nevyhýbaly využívání velevrubů infikovaných slávičkami a třikrát bylo pozorováno tření přímo do slávičky. Celkově se projevil pokles v reprodukčním úspěchu hořavky se vzrůstajícím množstvím sláviček přichycených na hostitelském velevrubu. Tato disertační práce ukázala, že různé nepůvodní populace mohou mít různý dopad na původní společenstva, stejně jako se původní populace mohou lišit v odezvě na různé populace nepůvodního druhu.

Abstract:

Invasions of non-native species threaten many native species, communities, and ecosystems. Anthropogenic factors can have a significant impact on contemporary distribution of intraspecific genetic diversity of non-native and native organisms. The presence of non-native species can affect interspecific coevolved associations within the communities of the native organisms. Interacting species often coexist in spatially structured populations that may coevolve at different rates, producing a geographic mosaic of adaptations and counter-adaptations. The objective of this study was to study population and population-genetic aspects of biological invasions on the host-parasite relationships used behavioural and molecular approaches. The study system used in this thesis was reciprocal relationship between the European bitterling (Rhodeus amarus) and their mussel hosts from family Unionidae influenced by recent invasions of the Chinese huge mussel (Sinanodonta woodiana) and the zebra mussel (Dreissena polymorpha). Molecular approaches have been used to estimate the genetic diversity and structure of the native populations of R. amarus and the introduced populations of S. woodiana. Behavioural methods have been used to study how invasion of D. polymorpha affects the microevolutionary processes of the oviposition decisions of R. amarus and their population consequences. The analysis of mitochondrial DNA of the bitterling fishes revealed six well-supported lineages, four of them are geographically restricted in Aegean, Caspian and Caucasian regions. This pattern is supported by the analysis of nuclear microsatellites, which showed small-scale differentiation of populations. Profound genetic structure among populations demonstrates the important role of genetic drift in the differentiation of the bitterling populations. The results showed the persistence of the European bitterlings in several small isolated populations during glacial. Only two lineages colonized most continental Europe from this refugial area with secondary contact in Western and Central Europe. Drainage boundaries have been undermined by human-mediated translocations, likely related to common carp cultivation and game fish management. Low mitochondrial diversity of the Chinese huge mussel was found in non-native populations in Europe. A single haplotype detected in all European samples matched the most common haplotype present in the central part of its natural distribution in China, pointing towards the potential primary source region. Analysis of microsatellite markers indicated a high level of admixture among European populations with intensive and ongoing gene flow. The Approximate Bayeasian Computation modeling showed complex associations among populations across the non-native European range There was no effect of recent sympatry between bitterling and zebra mussel on oviposition decisions and only weak effect on their reproductive success. Females inspected infested and non-infested mussels at the same rate but preferred to oviposit into non-infested unionid hosts. Females do not completely avoided oviposition into infested unionids and three ovipositions into zebra mussels were observed. Overall, there was a decrease in reproductive success of the bitterling fish with the increasing number of zebra mussels on unionid host shells. This dissertation thesis showed that different non-native populations can have different impacts on the native communities, as well as that the native populations can vary in response to different populations of non-native species.
