Phylogeny and system of European crayfish and their relation to other crayfish groups (Crustacea, Decapoda, Astacoida)

Abstract:

Bakalářská práce se zaměřuje na rozbor fylogenetických a systematických prací o sladkovodních racích. Pomocí rozpracované a odzkoušené metodiky zahrnující izolaci DNA, PCR a elektroforézu byly získány sekvence genu cytochrom c oxidázy podjednotky I. Druhy zařazené do této práce byly vybrány na základě čeledí a biogeografického rozšíření. Ve výsledné fylogenetické analýze se rod Cambaroides vyčlenil mimo čeleď Cambaridae, a obsadil bazální pozici v rámci Astacoidea. Rod Pacifastacus stojí bazálně k čeledi evropských druhů raků. Tyto výsledky jsou částečně v rozporu s obecně uznávanou taxonomickou klasifikací, podle které je rod Cambaroides v současnosti řazen do čeledi Cambaridae, stejně jako rod Pacifastacus do čeledi Astacidae. Získané výsledky jsou však v souladu s některými nedávnými molekulárními studiemi jiných autorů, a částečně i s analýzami založenými na morfologických znacích.

Abstract:

This thesis focuses on the analysis of phylogenetic and systematic papers on freshwater crayfish. Using the developed and tested methodology including DNA isolation, PCR and electrophoresis were obtained gene sequences of cytochrome c oxidase subunit I. Species included in this study were selected based on families and biogeographic distribution. In the resulting phylogenetic analysis was the genus Cambaroides earmarked outside the family Cambaridae and occupied a basal position within Astacoidea. Genus Pacifastacus, stands basally to the family of European crayfish species. These results do not fully support the generally accepted taxonomic classification, according to which the genus Cambaroides currently included in family Cambaridae, as well as the genus Pacifastacus in the family Astacidae. The results are consistent with some recent molecular studies of other authors, and partly with analyzes based on morphological characters.
