Abstract:

Makromolekuly kyseliny hyaluronové (HA) jsou lineárně nerozvětvené řetězce obsahující disociovatelné karboxylové skupiny (-COOH). Proto se tato kyselina ve vodných roztocích chová jako záporně nabitý polyelektrolyt a její konformace, tvar, rozměry i stupeň hydratace závisí na stupni disociace, který je určen pH a iontovou silou, přítomností kladně nabitých iontů, jejich povahou a mocenstvím. Ve vodných roztocích se polymerní řetězec HA nachází ve formě náhodně uspořádaného makromolekulárního klubka. Její chování však vykazuje určitý stupeň tuhosti způsobený systémem pevných vodíkových vazeb. Vzájemný kontakt mezi makromolekulárními řetězci indukuje vznik další terciární a vyšší struktury, která je způsobená superpozicí elektrostatických repulzí ovlivňovaných iontovou silou, nebo pH prostředí. Tato doktorská disertační práce je proto zaměřena na měření excitačních a emisních spekter roztoků fluoresceinů (fluorescein sodný, fluoresceinamin a fluorescein isothiokyanát-dextran), při různých koncentracích a vlnových délkách s využitím těchto fluorescenčních značek pro sledování konformačních změn kyseliny hyaluronové (sodné soli) a karboxymethylcelulózy. Měření se prováděla na fluorescenčním spektrofotometru Schimadzu RF-1501 (Japonsko). Přístroj měří fluorescenční spektra v rozsahu 220 nm až 900 nm a umožňuje automatické vyhledání optimální vlnové délky pro excitaci a emisi záření. Měření vzorků na fluorescenčním spektrofotometru sleduje závislost intenzity fluorescence na koncentraci rozpuštěné látky fluoroforu. Byla provedena fluorescenční měření v přítomnosti makromolekul kyseliny hyaluronové a karboxymethylcelulózy v závislosti na pH studovaných roztoků. Z naměřených hodnot byly sestaveny grafy závislosti intenzity exitačních a emisních spekter na pH prostředí pro oba studované polysacharidy. Tato měření umožnila sledování konformačních změn těchto aplikačně významných biopolymerů v závislosti na pH. Bylo zjištěno, že kalibrační přímky měřených koncentračních závislostí všech studovaných fluorescenčních značek bez přítomnosti biopolymeru vykazují lineární závislost na koncentraci, což umožňuje jejich praktické využití k analytickým účelům. Dále bylo zjištěno, že tyto lineární závislosti v přítomnosti studovaných biopolymerů (sodné soli kyseliny hyaluronové a karboxymetylcelulózy) mění charakter z jednoduché přímkové závislosti na dvojitou lineární závislost s průsečíkem indikujícím přechod expandované klubko/sbalené klubko při daném kritickém pH. Tato skutečnost umožní do budoucna konstrukci specifických čidel monitorujících konformační změny polysacharidů fluorescenčními měřeními pomocí bio-zobrazovacích sond.

Abstract:

The macromolecules of hyaluronid acid (HA) are linear unbranched chains containing dissociable carboxyl groups (-COOH). Therefore this acid behaves as a negatively charged polyelectrolyte in aqueous solutions and its conformation, shape, size and degree of hydration depends on the degree of dissociation, which is determined by pH and ionic strength, the presence of positively charged ions, their nature and valency. The polymer chain of HA is in the form of randomly organized macromolecular coil in aqueous solutions. Its behavior, however, shows a certain degree of rigidity caused by the system of solid hydrogen bonds. The mutual contact between the macromolecular chains induces more tertiary and higher structures, which is caused by the superposition of electrostatic repulsion influenced by the ionic strength or pH environment. This thesis is aimed on measuring the excitation and emission spectra of stock fluorescein solutions (e.g. fluorescein natrium, fluorescein amine and fluorescein isothiocyanate-dextran) in different concentrations and wavelengths using these to monitor the conformational change of the hyaluronic acid (sodium salt) and carboxymethylcelluloze. Measurement was performed by Schimadzu RF-1501 Fluorescence Spectrophotometer (Japan). This instrument measures fluorescence spectrums in the 220 nm to 900 nm wavelength range and can automatically find the optimal wavelength for the excitation and emission of radiation. Sample measurement on a fluorescence spectrophotometer monitored the dependence of fluorescence intensity on the concentration of a dissolved substance. Measurements of fluorescence were performed with the addition of hyaluronic acid and carboxymethylcellulose and adjusted by pH solutions using NaOH and HCl. Dependence charts intensity excitation and emission spectra on the pH were compiled from the measured values. These measurements allowed monitoring conformational changes of biopolymers depending on the pH It was found that the calibration curves of the measured concentration dependencies of all studied fluorescent labels without the presence of the biopolymer evince the linear dependence on the concentration, which allows their practical use for analytical purposes. Furthermore, it was found that these linear relationships change the nature of the simple linear dependence on double linear dependence with the intersection indicating a transition expanded coil / packed coil at the critical pH in the presence of studied biopolymers (sodium hyaluronate and carboxymethylcellulose). This fact allows a construction of the specific sensors monitoring conformational changes of the polysacharides with fluorescent measurements using bio-imaging probes for the future.
