Demonstration of gene Her-2/neu in mammary carcinoma

Abstract:

Rakovina prsu je nejčastějším nádorovým onemocněním u žen. Základem jejího úspěšného léčení je včasná diagnóza. V ČR je každoročně diagnostikováno více než 6 000 nových případů onemocnění, což je přibližně 120 pacientek na 100 000.Mezi příčinami úmrtí ve věkové kategorii 20–54 let zaujímá karcinom prsu první místo.Do základního diagnostického standartu byly zahrnuty výsledky vyšetření exprese hormonálních receptorů, proliferačních markerů a některých onkoproteinů. Bakalářská práce se zabývá problematikou onkoproteinů u karcinomů mléčné žlázy. Teoretická část zahrnuje popis genu HER-2/neu, jeho amplifikaci a objasňuje jeho roli při tomto onemocnění. Následuje popis karcinom prsu, rizikové faktory vzniku, jeho vyšetření, prognózu a typy léčby.Praktická část je zaměřena na přínos a srovnání imunohistochemického a molekulárně diagnostického vyšetření metodou FISH. Metodika je provedena na 50 tkáních formalínem fixovaných a v parafínu zalitých.

Abstract:

The breast cancer is the most common disease of women. The basis of successful treatment is early diagnosis. More than 6 000 new cases are diagnosed in the Czech Republic, which is approximately 120 patients for 100 000, every year.Among the causes of death in the age group of 20-54 year the breast cancer occupy the first place.In basic diagnostic standard were included the results of these examinations: expression of hormonal receptors, proliferation markers and some onkoproteins. This bachelor thesis deals with issue of onkoproteins of mammary carcinoma. The theoretical part includes description of gene HER-2/neu, its amplification and clarify its role in this disease. . It is followed by the description of the breast cancer, the risk factors for this desease, physical examination, prognosis and different types of therapy. The practical part is focused on benefist and comparison of immunohistochemical and molecular diagnostic tests by FISH method. The methodology is conducted on formalin-fixed and paraffin-embedded breast cancer tissues.
