Extraction of photosynthetically active pigments using small volume method

Abstract:

Tato bakalářská práce je zaměřena na extrakci fotosyntetických pigmentů z rostlinného materiálu pomocí metody malých objemů. V teoretické části jsou obsaženy informace o funkci, fyzikálně-chemických vlastnostech, extrakci a kvantitativní analýze fotosyntetických pigmentů. V rámci experimentální části byla zavedena maloobjemová extrakční metoda a porovnána se standardní extrakční metodou, která se na Katedře fyziky PřF OU používá. Za účelem ověření univerzálnosti maloobjemové extrakční metody se pracovalo s šesti rostlinnými druhy. Obsah fotosyntetických pigmentů v extraktech byl stanoven spektrofotometricky. Výtěžky obou extrakčních metod byly statisticky srovnány. Výsledky prokázaly, že maloobjemová extrakční metoda vykazuje při extrakci fotosyntetických pigmentů z listů/jehlic vyšších rostlin stejnou výtěžnost jako metoda standardní, a lze ji tedy použít i v případě omezeného množství rostlinného materiálu.

Abstract:

This bachelor thesis is focused on the extraction of photosynthetic pigments from plant material using small-volume method. The theoretical part contains information about the function, physical-chemical properties, extraction and quantitative analysis of photosynthetic pigments. In the experimental part, the small-volume extraction method was introduced and compared with the standard extraction method used at the Department of Physics of the University of Ostrava. In order to verify the versatility of the small-volume extraction method, six plant species were used. The content of photosynthetic pigments in extracts was determined by spectrophotometric method. The yields of both extraction methods were statistically compared. The results revealed that the small-volume extraction method has the same yield as the standard extraction method for the extraction of the photosynthetic pigments of the leaves/needles of higher plants. Therefore it is possible to use it when the amount of plant material is limited.
