Characterization of neutrophil cell death associated with oxidative burst

Abstract:

Zánětlivá reakce nastupuje jako odpověď na porušení integrity organismu. Důleţitou roli v ní hrají neutrofilní granulocyty, které jsou během zánětu aktivovány a pronikají do zanícené tkáně. Avšak masivní akumulace neutrofilních granulocytů v místě zánětu může negativně ovlivnit procesy hojení a iniciovat patologické zánětlivé procesy. Takže účinné odstraňování neutrofilních granulocytů zprostředkované indukcí regulované buněčné smrti je klíčovým procesem bránící rozšiřování těchto patologických podmínek. Myeloperoxidáza (MPO), hojně se vyskytující enzym v granulích neutrofilů, primárně spojována s obranou hostitele, by mohla hrát významnou roli v regulované buněčné smrti neutrofilních granulocytů. Avšak význam MPO v mechanismu buněčné smrti neutrofilních granulocytů nebyl dosud objasněn. Tato bakalářská práce se zabývá charakterizací buněčné smrti neutrofilních granulocytů spojené s oxidativním vzplanutím. V předchozích studiích bylo zjištěno, že dochází k expresi fosfatidylserinu, typického znaku apoptózy, na povrchu neutrofilních granulocytů. Tato exprese je však snížená u MPO deficientních neutrofilů. Avšak další markery typické pro apoptózu (aktivace kaspáz, DNA fragmentace) nebyly pozorovány. Naproti tomu bylo detekováno štěpení proteinu s lehkým řetězcem 3 a aktivace proteinu p62, typických autofagických markerů, avšak bez rozdílu mezi kontrolními a deficientními neutrofily. Stejně tak aktivace receptor-interagující serin/threonin protein-kinázy 1, proteinu typického pro nekroptózu, byla pozorována u kontrolních a MPO deficientních neutrofilních granulocytů bez významnějších rozdílů. Je tedy potřeba pokračovat v charakterizaci vlivu MPO na životnost neutrofilních granulocytů.

Abstract:

Inflammation is a response of organism to interruption of its integrity. Neutrophil granulocytes play an important role in this process. They are activated during the inflammation and penetrate to the inflamed tissue. However a massive accumulation of neutrophil granulocytes in the site of inflammation can negatively influence processes of healing and iniciate pathological inflammatory conditions. So effective removal of neutrophil granulocytes that is performed by induction of regulated cell death is crucial process that protects the organism from a propagation of these inflammatory conditions. Myeloperoxidase (MPO) is the most abundant enzyme contained in granules of neutrophils and it is primarily associated with a host defence. MPO could play an important role in a regulated cell death of neutrophil granulocytes. But its importance in this process has not been elucidated yet. This bachelor’s thesis deals with the characterization of neutrophil cell death associated with an oxidative burst. The expression of phosphatidylserine, a typical marker of apoptosis, was observed on neutrophil surface in previous studies. Nevertheless this expression is lower on a surface of MPO deficient neutrophils. Another markers of apoptosis (activation of caspases, DNA fragmentation) were not detected. In contrast the cleavage of protein light chain 3 and activation of protein p62, typical proteins of autophagy, were observed. But there is not any difference between control and MPO deficient neutrophils. Activation of protein receptor-interacting serine/threonine-protein kinase 1, typical marker of necroptosis, was also detected but as well as in the case of autophagic markers there is not any significant difference between control and MPO deficient neutrophils. It is necessary to continue with the characterization of MPO influence to neutrophil’s viability.
