Abstract:

Shrnutí WT-1 peptid byl pro prvotní experimenty studie peptidových protinádorových vakcín na UCBI vybrán především díky jeho specifickým, pro protinádorové imunomodulace optimálním, vlastnostem. Jeho výskyt je zaznamenán u řady nádorových onemocnění, počínaje solidními nádory plic, prsu, vaječníků, štítné žlázy, tlustého střeva a konečníku a konče leukemiemi. Exprese peptidu WT-1 za fyziologických podmínek je navíc zaznamenána pouze na buňkách fetálních jater, případně na buňkách varlat a vaječníků, tzn. v oblasti imunoprivilegovaných orgánů. Tyto skutečnosti naznačují, že při zaměření imunoterapeutické vakcinace proti peptidu WT-1 by měly být zaznamenatelné minimální interference vedoucí k nežádoucím vedlejším účinkům. Cílem této práce bylo především seznámení s metodickými postupy, které se nejvíce využívají při stanovení přítomnosti a imunogenicity vybraného nádorového antigenu. Praktickou částí je poté doplněno vlastní provedení vybraných experimentů. Konečné vyhodnocení úspěšnosti vyvolání a detekce komplexní T lymfocytární odpovědi na přítomnost synteticky připravených peptidů WT-1124-138 a WT-1126-134 bylo provedeno u dárce, jehož reaktivní profil alespoň z části odpovídal profilu očekávanému. Z řady metod použitých pro vyhodnocení reaktivity T lymfocytů vybraného dárce se jako nejprůkaznější jeví detekce povrchové produkce IFN-γ, kdy byla po 48 a 72 hodinách od zahájení kultivace zaznamenána zvýšená produkce tohoto cytokinu Th lymfocyty stimulovanými 500nM i 5000nM peptidem WT-1126-134. Tato aktivace byla navíc částečně potvrzena metodou detekce povrchové exprese molekuly CD25. Vybrané experimenty však dosud nebyly provedeny na statisticky hodnotitelném množství dárců, proto nelze zcela zavrhnout ani další prováděné metody – detekce proliferace buněk barvených CFSE či detekce hladiny vybraných cytokinů. Jejich zařazení do budoucích, nově plánovaných experimentů, bude dozajista velmi přínosné.

Abstract:

Summary Two immunodominant epitopes derived from WT-1 antigen were chosen for the first experiments in the research on peptide cancer vaccines, which are practised at the University Cell Immunotherapy Center, because of their special features optional for immunotherapy. Occurence of the WT-1 peptide is known in diverse human tumors, including lung cancer, breast cancer, ovarian cancer, thyroid cancer, colorectal cancer and leukemia. During normal ontogenesis the WT-1 peptide occurs only in fetal kidney, ovary and testis. The surface of these vitals isn´t recognized by the immune system, so that the presence of any side effects caused by application of the WT-1 vaccines should be minimal. It is the reason for using the peptides derived from WT-1 as a new purpose in immunotherapy. The main purpose of this study was to refer some methods to determine the presence and the immunological abilities of the chosen cancer antigen. The study contains a number of results from the first experiments with the WT-1 peptide. The final evaluation of the experiments with the WT-1124-138 and WT-1126-134 peptides was realized on one blood donor. The most demonstrable method seems to be an IFN-γ Secretion Assay. Through the use of this method the activation of Th lymphocytes, which were stimulated with 500nM and 5000nM of WT-1126-134 peptide, was found. The activation, detected after 48 and 72 hours from the biginning of the cultivation, was reconfirmed through the use of the metod which detects the expression of the CD25 molecule. But the number of practised experiments wasn´t statistically relevant, so that the other methods, for example the detection of the proliferation and the detection of the presence of some cytokines, should be certainly included in the future experiments with cancer peptides.
