Capsid-binding inhibitors of honeybee viruses from families Dicistroviridae and Iflaviridae

Abstract:

Dicistroviry a iflaviry jsou malé ikosaedrální viry, které snižují produktivitu i životaschopnost včelstev. V současné době není k dispozici žádné léčivo proti těmto virům. Pomocí sekvenční a strukturní analýzy (za využití modelu sestaveného dle sekvence) s podobnými picornaviry je možné odhadnout účinek picornavirových inhibitorů proti dicistrovirům a iflavirům. Tato práce popisuje včelí dicistroviry a iflaviry, jejich strukturu, životní cyklus, šíření virové infekce a možnosti léčby pomocí kapsid-vazebných inhibitorů. Sekvenční a strukturní analýza ukázala, že u vybraných včelích virů se pravděpodobně nenachází vazebné místo pro kapsid-vazebné inhibitory. Pro potvrzení této teorie je ale nutné znát přesnou strukturu ne pouze modelovou, která byla použita v této práci. Experimentální část se zabývá klonováním a purifikací proteasy včelího viru, což umožní vývoj inhibitorů proteasy.

Abstract:

Dicistroviruses and iflaviruses are small icosahedral viruses, which decrease productivity and viability of honeybee colonies. To date there is no drug available against these viruses. Thanks to sequence and structural (using the models predicted based on sequence) comparison with picornaviruses it is possible to predict the effect of inhibitors on dicistroviruses and iflaviruses. This thesis describes honeybee dicistroviruses and iflaviruses, their structure, lifecycle, transmission modes and possibilities of cure by capsid-binding inhibitiors. Sequence and structure analysis showed, that honeybee viruses probably do not contain binding site for the capsid-binding inhibitors. For confirmation of this theory it is necessary to know the real structures, not just the models, which were used in this thesis. Experimental part deals with cloning and purification of protease of one of honey bee viruses, which will allow the development of protease inhibitors.
