The effect of gregarines on survival and development of laboratory reared mealworm beetles

Abstract:

Bakalářská práce se skládá z teoretické a praktické části. Teoretická část této bakalářské práce se věnuje charakteristice potemníka moučného (Tenebrio molitor) a charakteristice jednotlivých druhů gregarin, které jej parazitují. Zejména se jedná o druhy Gregarina cuneata, Gregarina polymorpha a Gregarina steini, které parazitují larvální stádia potemníků. Často najdeme v jedné larvě právě všechny tyto druhy gregarin. Rešerše je zároveň zaměřena na hostitelsko-parazitické vztahy a pokouší se shrnout, jakým způsobem je potemník moučný gregarinami ovlivňován. Praktická část popisuje a diskutuje provedené experimenty. Prvním z nich je čištění larev potemníků od nákazy gregarinami, které bylo ve všech případech úspěšné. Dalším experimentem je sběr gametocyst a jejich excystace. Během vícerých pokusů se vyskytly překážky, přesto byl nakonec i tento experiment úspěšný. Poslední experiment spočíval v experimentální nákaze vyčištěných larev potemníků vybraným druhem gregariny. Provedeny byly celkem 3 pokusy a v posledním z nich bylo dosaženo úspěchu. Ze všech tří experimentů se však tento ukázal jako nejvíce problematický. Poslední část diskuse na základě těchto experimentů zároveň navrhuje možnosti dalšího bádání, které by nás mohlo obohatit o další informace týkající se hostitelsko-parazitických interakcí mezi potemníky a gregarinami.

Abstract:

The bachelor thesis comprises the theoretical and practical part. The theoretical part of this thesis provides information about the yellow mealworm (Tenebrio molitor) and describes general characteristics of its gregarine parasites, Gregarina cuneata, Gregarina polymorpha and Gregarina steini parasitizing the larval stages of mealworm beetle. Often all these gregarine species can be found within a single host. The literary research also focuses on the host-parasite interactions and impact of gregarines on the yellow mealworm. The practical part describes and discuses performed experiments. The first one tested the sterilization of the yellow mealworms of gregarine infection and all trials were successful. The second experiment was based on gametocysts’ collection and excystation. Though some problems emerged during first trials, in the end the experiment was successful. The aim of the last experiment was to re-infect the gregarine-free mealworms with selected species of Gregarina. Three trials were carried out and only the last one was successful. This experiment showed to be the most complicated. Based on experiments mentioned above, the last part of discussion also suggests the possibilities for further research that could provide more information on the host-parasite interactions between yellow mealworms and gregarines.
