Separation of pigment-protein complexes using CN-PAGE with gradient gels

Abstract:

Tato bakalářská práce je zaměřena na možnosti separace pigment-proteinových komplexů (PPC) z thylakoidních membrán chloroplastů vyšších rostlin pomocí nativních elektroforetických technik. V první části se věnuji popisu složení a funkce hlavních PPC ? fotosystému II (PSII) a fotosystému I (PSI), popisu nativní elektroforézy obecně a samotné metodě CN-PAGE (bezbarvé nativní elektroforézy; ?colourless? nebo také ?clear native PAGE?). Druhou část práce věnuji popisu vybraných publikací jiných autorů, zabývajících se separací PPC pomocí nativních elektroforetických technik (CN-PAGE, zelené elektroforézy a Deriphatové elektroforézy). Zpracovány jsou práce, zabývající se elektroforetickou separací PPC pomocí gelů jak s gradientem koncentrace akrylamidu a bisakrylamidu, tak také pomocí jednokoncentračních polyakrylamidových gelů. Pro dosažení šetrné a co nejlepší separace PPC se jeví jako nejvhodnější použití dodecyl maltosidu (DM) jako solubilizačního detergentu a kombinace detergentů DM+DOC (deoxycholátu sodného) přidávaných do elektroforetických pufrů. Použitím kombinace těchto detergentů je dosaženo separace superkomplexů fotosystému I (PSI) a fotosystému II (PSII), fotosystému I (PSI), jádra fotosystému I (CCI) a fotosystému II (CCII), světlosběrných komplexů PSII v trimerním a monomerním stavu (LHCIIt a LHCIIm).

Abstract:

This thesis is focused on possibilities of separation of pigment-protein complexes (PPCs) from thylakoid membranes of higher plant chloroplasts using native electrophoretic techniques. The first part of the thesis is devoted to description of the composition and function of the major PPCs ? photosystem II (PSII) and photosystem I (PSI), to general description of the native electrophoresis principle and CN-PAGE itself (?colourless? native electrophoresis or ?clear native PAGE?). The second part is focused on the description of selected publications dealing with PPC separation by native electrophoretic techniques (CN-PAGE, green electrophoresis and Deriphat electrophoresis). Papers dealing with the separation of PPCs using gels with gradient of total acrylamide and bisacrylamide concentration and with uniform concentration gels are described. To achieve a mild and the best possible PPC separation by native PAGE the using of dodecyl maltoside (DM) as a solubilization detergent and combination of detergents DM+DOC (sodium deoxycholate) added to the electrode buffers seems to be optimal. Using a combination of these detergents a separation of supercomplexes of photosystem I (PSI) and photosystem II (PSII), photosystem I (PSI), core complexes of PSI (CCI) and PSII (CCII), light-harvesting complexes of PSII in trimeric and monomeric form (LHCIIt and LHCIIm) is achieved.
