Genotypization of group G streptococci using polymerase chain reaction

Abstract:

V rámci předložené diplomové práce jsme zavedli druhově specifickou polymerázovou řetězovou reakci pro druhovou identifikaci kmenů streptokoků se skupinovým antigenem G izolovaných z klinického materiálu humánní provenience. Dále jsme se zabývali detekcí genu kódujícího streptolyzin S, a to rovněž s využitím polymerázové řetězové reakce. Provedli jsme optimalizaci polymerázové řetězové reakce pro průkaz Streptococcus (S.) canis s využitím primerů cI a cII. Detekční limit této reakce jsme stanovili na 130 pg DNA. Dále jsme pak zavedli zcela novou metodu polymerázové řetězové reakce pro identifikaci S. dysgalactiae ssp. equisimilis. Detekčním limitem této reakce je 1,4 ng DNA. Celkem jsme zpracovali 141 kmenů streptokoků se skupinovým antigenem G získaných od lidí z různého klinického materiálu. DNA jsme izolovali fenol-chloroformovou extrakcí z 24-hodinové kultury pomnožené v Todd-Hewitově bujónu. U 133 kmenů jsme pomocí druhově specifické PCR prokázali druh S. dysgalactiae ssp. equisimilis. Žádný z kmenů nebyl pomocí PCR identifikován jako S. canis. Zbývajících 8 kmenů nebylo možné zařadit ani do jednoho z těchto dvou druhů. Naproti tomu bylo na základě fenotypových vlastností 119 kmenů identifikováno jako S. dysgalactiae ssp. equisimilis, 8 kmenů jako S. canis a 2 kmeny jako S. uberis. Zbývajících 12 kmenů se podařilo zařadit pouze mezi streptokoky se skupinovým antigenem G. Rozdíly mezi výsledky mikrobiologického vyšetření a výsledky polymerázové řetězové reakce mohou být způsobeny variabilitou fenotypových vlastností vyšetřovaných kmenů, které nemusí přesně vždy odpovídat standardním vlastnostem uváděným pro jednotlivé druhy. V neposlední řadě jsme zavedli polymerázovou řetězovou reakci pro průkaz genu kódujícího streptolyzin S s využitím primerů SI a SII. Detekční limit této reakce jsme stanovili na 1,6 ng DNA.

Abstract:

The aim of presented thesis was the development of the species-specific polymerase chain reaction for the identification of group G streptococci isolated from the clinical material of human origin. The second part of the work dealt with the detection of the gene encoding streptolysin S using the polymerase chain reaction. We performed an optimization of polymerase chain reaction for detection of Streptococcus (S.) canis using primers cI and cII. The detection limit of this reaction was determined at 130 pg DNA. Further, we introduced a completely new method of polymerase chain reaction for identification of S. dysgalactiae ssp. equisimilis with the detection limit of 1,4 ng DNA. Altogether 141 strains of group G streptococci isolated from various clinical specimens of human origin were processed. The DNA was isolated by the phenol-chloroform extraction from the 24-hour bacterial broth culture. Out of total 141 GGS strains, 133 strains were identified as S. dysgalactiae ssp. equisimilis using the species-specific polymerase chain reaction. None of the strains was identified as S. canis. The remaining 8 strains could not be assigned to either of these two species. In contrast to these results, 119 strains were identified as S. dysgalactiae ssp. equisimilis, 8 strains as S. canis and 2 strains as S. uberis according to their phenotypic properties. The remaining 12 strains were classified only as group G streptococci. The differences between the results of the microbiological investigation and the results determined by the species-specific polymerase chain reaction may be caused by the variation in the phenotypic properties of tested strains, that don't always correspond to the standard properties given for individual species. Furthermore, we introduced the polymerase chain reaction for the detection of sagA, the gene encoding streptolysin S, using primers SI and SII. The detection limit was determined at 1,6 ng DNA.
