Identification of methanogenic Archaea in antarctic lake sediment

Abstract:

Metanogenní archea jsou producenty metanu v mnoha anaerobních prostředích. Tento skleníkový plyn se podílí na neblahém jevu globálního oteplování, ale zároveň může být využit jako palivo v podobě bioplynu. Jezera a jejich sedimenty představují nezanedba-telnou součást výskytu metanogenů, s čímž souvisejí i emise metanu do ovzduší. Antarktická jezera jsou zatím málo prostudována, co se týká aktivity a složení těchto mikroorganismů, ačkoli prostředí poskytuje specifické podmínky, které nelze najít nikde jinde na světě. V této práci byly zpracovávány vzorky sedimentů sladkovodních antarktických jezer na ostrově James Ross a ostrovech Long Island a Vega v jeho okolí. Pro kvantifikaci a přítomnost metanogenů bylo využito amplifikace funkčního genu mcrA metodou qPCR. Následné taxonomické složení bylo získáno sekvenací stejného genu metodou NGS. Sekvenován byl i gen pro 16S rRNA k zjištění role metanogenů v rámci prokaryotické komunity. Dominantními se ukázaly kmeny Proteobacteria, Bacteroidetes, Verrucomicrobia, Chloroflexi, Acidobacteria a Actinobacteria, které se převážně podílejí na funkcích aerobní chemoheterotrofie, fermentace, fototrofie, nitrifikace, redukce nitrátů a respirace sirných sloučenin. Metanogeny představují minoritní část v celkovém složení prokaryot a jejich četnost se pohybuje v rozsahu 10^1-10^6 na gram sedimentu. Přítomní byli zástupci od všech tří typů metanogeneze s převažujícími rody Methanothrix a Methanobacterium. Byla demonstrována i přítomnost potenciálně termofilních zástupců. Vliv parametrů prostředí však nebyl prokázán. Tento fakt společně s velkým podílem blíže neklasifikovatelných sekvencí z obou sekvenačních běhů naznačuje, že jezera v okolí ostrova James Ross se jeví jako bohatý zdroj příležitostí pro další studie metanogenů a jiné mikrobiologické výzkumy.

Abstract:

Methanogenic archaea are producers of methane in many anaerobic environments. This greenhouse gas contributes to the unfortunate phenomenon of global warming. On the other hand, it can be used as a fuel in the form of biogas. Lakes and their sediments form a significant habitat for methanogens which is connected to methane emissions into the air. Antarctic lakes are so far little studied regarding activities and composition of these microorganisms, although the environment provides conditions that can not be found anywhere else in the world. In this work, sediment samples of freshwater Antarctic lakes on James Ross Island and Long Island and Vega Island in its vicinity were processed. Amplification of the functional mcrA gene by qPCR was used for quantification and the presence of methanogens. The following taxonomic composition was performed by NGS sequencing of the same gene. The 16S rRNA gene was also sequenced to determine the role of methanogens within the prokaryotic community. Predominant phyla are Proteobacteria, Bacteroidetes, Verrucomicrobia, Chloroflexi, Acidobacteria and Actinobacteria, which are mainly involved in aerobic chemoheterotrophy, fermentation, phototrophy, nitrification, nitrate reduction and respiration of sulfur compounds. Methanogens make up a minor part of the total composition of prokaryotes, and their abundance is in the range of 10^1-10^6 per gram of sediment. Representatives of all three types of methanogenesis are present with the predominant genera Methanothrix and Methanobacterium. The presence of potentially thermophilic representatives has also been demonstrated. The influence of environmental parameters has not been proven. Together with a large proportion of unclassified sequences from both sequencing runs, this fact indicates that lakes around James Ross Island are a promising rich source of opportunities for further studies of methanogens and other microbiological researches.
