Modern Therapy of Solid Tumors Using Ferritin Nanoparticles

Abstract:

Využití nanoterapeutik v terapii nádorů a rozvoj nanomedicíny byl podpořen zejména použitím nanočástic pro cílenou dopravu léčiv do nádoru. Nicméně mezi nejčastější léčebný postup stále patří chemoterapie. Hlavním nedostatkem tohoto léčebného přístupu je fakt, že kromě rychle se dělících nádorových buněk ovlivňuje i zdravé buňky. Tato diplomová práce se zaměřuje na využití feritinových nanočástic obsahujících železo (Ftn-Fe) v nádorové terapii. Cytotoxicita Ftn-Fe byla nejprve otestována na myších (4T1, B16F10, CT26) a lidských (DU145, MCF-7, MDA MB 468, SW480) nádorových buněčných liniích pomocí MTT assay. Na všech použitých buňkách vykazoval Ftn-Fe cytotoxicitu, vyjma B16F10 buněk, s hodnotami IC50 v rozmezí od 0,15 mg/ml do 0,39 mg/ml. Na základě výsledků se práce dále zaměřuje na typ buněčné smrti indukované Ftn-Fe. V závislosti na koncentraci Ftn-Fe indukoval ferroptózu na buňkách 4T1. Navíc byl pozorován synergismus mezi cytotoxickými účinky Ftn-Fe a Dox. Závěr praktické části práce je zaměřen na optimalizací syntézy feritinu s definovaným množstvím železa, kde byl testován poměr Fe2+ roztoku a ddH2O. Jako nejvhodnější byl stanoven poměr 1:6. Na základě těchto experimentů bylo potvrzeno potenciální využití feritinových nanočástic pro terapii solidních nádorů.

Abstract:

The recent progress in nanomedicine has become the new field of cancer therapy mostly based on drug delivery systems. However, chemotherapy still represents the most common treatments strategy for solid tumors. The lack of selectivity is the main drawback of using chemotherapy for tumor treatment. Thus, this diploma thesis is focusing on using Ftn-Fe nanoparticles in tumor treatment. First, the cytotoxicity of Ftn Fe was tested in mouse (4T1, B16F10, CT26) and human (DU145, MCF-7, MDA MB-468, SW480) tumor cell lines. Ftn-Fe showed significant cytotoxicity in all human and mouse cell lines except B16F10 with IC50 values ranging from 0.15 mg/ml to 0.39 mg/ml. According to cytotoxicity results, in the next step thesis was focused on cell death induced by Ftn-Fe in 4T1 cells. Ftn-Fe was able to induce ferroptosis in dose dependent manner in mouse 4T1 cell line. Moreover, Ftn-Fe showed synergism in cytotoxic activity with Dox. In conclusion, the practical part of diploma thesis is focusing on ferritin synthesis with define iron amount inside the ferritin cavity. In the protocol there was optimized the ratio between Fe2+ solution and ddH2O. The most effective with the highest iron amount inside the ferritin cavity was ration 1:6. Based on these experiments the potential use of ferritin nanoparticles in the treatment of solid tumors was confirmed.
