Assessment of genotoxic potency and risks of compounds associated with the suspended particulate matter in the air

Abstract:

Znečištění ovzduší je dobře známo svými škodlivými účinky na lidské zdraví včetně mutagenity a karcinogenity.Významný podíl na těchto účincích mají organické látky vázané na suspendované částice v ovzduší. Tvoří zde velice složitou směs, kterou nelze plně charakterizovat současně používanými chemickými metodami. Navíc, vhodná toxikologická data pro zhodnocení jejich genotoxického potenciálu a možných rizik jsou velmi omezená. Avšak je vyvíjena snaha najít vhodný způsob pro komplexní hodnocení znečištění ovzduší a jeho rizik. Jednu z možností představuje použití biotestů. V rámci této studie, byla zkoumána genotoxická aktivita organických polutantů jak vázaných v částicích tak v plynné fázi s využitím skríningového testu genotoxicity, SOS chromotestu, a to jak bez tak i s metabolickou aktivací. Na jednu stranu byla hodnocena aktivita organických látek vázaných na různé velikosti částic a na druhou stranu byl hodnocen příspěvek jednotlivých frakcí surového organického extraktu z částic na běžně pozorovanou genotoxickou aktivitu organických extraktů těchto částic. I když je převážná část aktivity spojena s polutanty vázanými na jemné částice, zejména v případě přímé genotoxicity, se mohou významně podílet na celkové aktivitě i látky vázané v hrubších frakcích částic. Přímá i nepřímá genotoxicita je spíše spojena s přítomností středně polárních až polárních látek než s neutrálními látkami jako jsou polycyklické aromatické uhlovodíky (PAHs). Na základě porovnání genotoxické aktivity semivolatilních organických látek v plynné fázi s aktivitou polutantů vázaných na částice bylo potvrzeno, že většina genotoxického potenciálu je spojena s látkami vázanými na částice, ale výsledky také částečně ukázaly, že plynné polutanty mohou potenciovat aktivitu polutantů vázaných na částice a proto by neměly být opomíjeny. Zjištěná genotoxicita byla také diskutována s ohledem na koncentrace PAH zjištěné v ovzduší, na jejichž základě je prováděno hodnocení možných zdravotních rizik organického znečištění ovzduší. Vedle ověření použitelnosti SOS chromotestu jako vhodného nástroje pro kvantifikaci genotoxického potenciálu organického znečištění ovzduší a identifikaci možných zdravotních rizik, bylo navrženo hodnocení možných genotoxických rizik na základě porovnání genotoxických potenciálů zjištěných pomocí biotestů. Poněvadž toxikologické informace pro mnoho nových prioritních perzistentních organických polutantů, tzv. nové environmentální polutanty, jsou dosud omezené pro účely hodnocení jejich zdravotních rizik, genotoxický potenciál N-heterocyklických polyaromatických uhlovodíků (NPAHs), obsažených v emisích ze spalování fosilních paliv a zjištěných v ovzduší, a 4,4’-difenochinonů a 4,4’-dihydroxybifenylů, zjištěných v popílku ze spalovny, byl zjišťován navíc v rámci této práce s využitím SOS chromotestu. 4,4’-difenochinony a 4,4’-dihydroxybifenyly vykázaly pouze slabou nepřímou genotoxickou aktivitu. Na druhou stranu, některé NPAHs jsou silně genotoxické. Na rozdíl od homocyklických PAHs, vykazují i přímou genotoxicitu. Tato aktivita byla zjištěna i u nízkomolekulárních zástupců (se 2 a 3 kruhy). Výskyt NPAHs v životním prostředí může představovat genotoxické riziko. Vzhledem k tomu, že obě skupiny testovaných látek se mohou vyskytovat v ovzduší, mohou přispívat částečně ke genotoxickému potenciálu organického znečistění ovzduší.

Abstract:

Air pollution is well-known for its adverse health effects including mutagenicity and carcinogenicity. A significant portion of the effects is related with the occurrence of organic compounds associated with the suspended particulate matter in the air. They form a very complex mixture which cannot be fully characterized by currently used chemical methods. Moreover, toxicological data that could be used for the evaluation of genotoxic potency and risks of such mixtures are very limited. However, there are efforts to find a suitable way how to evaluate the air pollution and its health risks in a complex way. Bioassays seem to be an appropriate tool for this purpose. In this study, genotoxic activities of the organic pollutants bound to the particulate matter as well as present as gases in the atmosphere were investigated using a screening genotoxicity test, SOS chromotest, both without and with metabolic activation. On one hand, the activity of organic pollutants in different size fractions of the particulate matter was determined and evaluated and, on the other hand, contributions of individual fractions of crude organic extracts of the particulate matter were evaluated to reveal what is behind the observed genotoxic potency of routinely analyzed organic extracts of the particulate matter. Even if the genotoxic activity is induced mainly by pollutants associated with fine mode particles, especially in the case of direct genotoxicity, the pollutants in coarse mode particles may also contribute significantly to the total activity. Both the direct and indirect genotoxic activity is related with moderately polar and polar organic compounds presence in the extracts rather than with neutral compounds like polycyclic aromatic hydrocarbons (PAHs). Based on comparison of the genotoxic activity of semi-volatile organic compounds in the gas phase the activity of pollutants absorbed to the particles, it was confirmed that majority of the genotoxic potency of the organic air pollution is related with the pollutants associated with the particulate matter, but the gaseous pollutants should be also considered, because they may potentiate the activity of the pollutants bound to the particles. The observed genotoxic potencies were also discussed in relation to PAHs levels detected in the air, routinely evaluated in relation to potential health risks of the organic air pollution. Besides confirmation of applicability of the SOS chromotest as a suitable tool for the quantification of genotoxic potency of the organic air pollution and identification of potential health risks, an approach of comparative risk assessment for the evaluation of potential genotoxic risks of the organic air pollution, based on the genotoxic potencies determined bioassays, has been proposed. Since toxicological information about a lot of persistent organic pollutants of new concern, so-called new environmental pollutants, has been so far limited for the evaluation of their health risks, genotoxic potencies of N-heterocyclic polyaromatic hydrocarbons (NPAHs), contained in emissions from combustion of fossil fuels and detected in the air, and 4,4’-diphenoquinones and 4,4’-dihydroxybiphenyls, detected in fly ash emitted from a waste incineration plant, were evaluated, in addition, within this study using the SOS chromotest. The 4,4’-diphenoquinones and 4,4’-dihydroxybiphenyls showed only a weak indirect genotoxicity in the SOS chromotest. On the other hand, some NPAHs were very potent genotoxicants. Contrary to homocyclic PAHs, they also showed direct genotoxic activity. The activity was even observed for low molecular weight NPAHs (2- and 3-ringed NPAHs). The occurrence of NPAHs in the environment may be genotoxic risk. Since both groups of the tested compounds may occur in the air, they can contribute partially to the genotoxic activity of the organic air pollution.
