Vývoj metod fenotypizace kořenů u polních plodin a vyhodnocení dopadu environmentálních stresů a výživy rostlin na architekturu kořenů

Abstract:

Předložená disertační práce se zabývá studiem vlivu environmentálních faktorů (atmosférické koncentrace CO2, UV záření a stresu sucha) a výživy N na nadzemní biomasu, výnos zrna a růst kořenů u ozimé pšenice a jarního ječmene a věnuje se také vývoji metod fenotypizaci kořenů u polních plodin. Výsledky práce ukazují, že zvýšení koncentrace CO2, vyloučení UV záření, aplikace N a stress sucha mají průkazný vliv na biomasu, výnos zrna a růst kořenů. Mimo to byly také zjištěny interakce mezi těmito faktory. Druhým cílem práce bylo otestovat funkčnost nově vyvinutého systému pro fenotypizaci kořenů pro vyhodnocení rozdílů v architektuře kořenového systému v rámci dvou experimentů, které byly zaměřeny na vliv nedostatku živin (N, P, a K) a spektrálního složení světla (UV-A, modré, červené a dlouhovlnné červené záření). Tyto experimenty prokázaly, že systém umožňuje snadný a opakovatelný přístup ke kořenům pro jejich měření a vzorkování. Vyvinutý fenotypizační systém představuje potenciál pro studium architektury kořenového systému s použitím obou testovaných softwarových nástrojů tj. SmartRoot a WinRHIZO.

Abstract:

The presented dissertation studies the effect of environmental factors (atmospheric CO2 concentration, UV radiation and drought stress) and N nutrition on above-ground biomass, grain yield and root growth of winter wheat and spring barley, and also tries to develop the methods for root phenotyping in field crops. The results showed that CO2 enrichment, UV exclusion, N application and drought stress had effect on biomass, grain yield and root growth. Moreover, the interactive effects between these factors were also found. The second aim is to verify the functionality of a newly developed root phenotyping system for evaluation of differences in root system architecture (RSA) within the two experiments, which were the effect of nutrient (N, P and K) deficiency and light spectral quality (UVA, Blue, Red and Far red light) on RSA. These experiments showed that this system allowed easy repeated non- invasive access to roots for their measuring and sampling. Our root phenotyping system offers the potential to study root system architecture and could be analysed by both software SmartRoot and WinRHZO.
