Searching for lectins in mushroom using bioinformatics

Abstract:

Lektiny jsou proteiny neimunitního původu, jejichž hlavní podstatou je schopnost velmi specificky a reverzibilně vázat sacharidy. Díky této vlastnosti se lektiny podílí na celé řadě fyziologických a patologických procesů. Tyto proteiny jsou schopny rozpoznat i nepatrné rozdíly v sacharidové struktuře. Jelikož nevykazují enzymatickou aktivitu, zůstává vazebný partner nezměněn. Všechny tyto vlastnosti činí z lektinů zajímavé objekty pro biomedicinský a biotechnologický výzkum. I když se lektiny v houbách vyskytují se stejnou hojností jako v ostatních organismech, jejich výzkumu se pozornosti dostává až v poslední době. Důvodem je nejspíš nedávno objevený antiproliferativní a imunostimulační potenciál těchto lektinů. Tato bakalářská práce se zabývá potenciálními lektiny z Agaricus bisporus. V první části se práce věnuje bioinformatické analýze sekvencí hypotetických lektinů získaných předchozím laboratorním výzkumem. Součástí je i predikce sktruktury nejslibnějších proteinů. Druhá část je zaměřena na laboratorní vyhledávání a izolaci lektinů z Agaricus bisporus var. brunensis pomocí afinitní chromatografie a ověření přítomnosti „ricin-B-like“ hypotetického lektinu v této variantě žampionu.

Abstract:

Lectins are proteins of non-immune origin, the main being of which is the ability to binding carbohydrates very specifically and reversibly. Thanks to this property, lectins are involved in a wide range of physiological and pathological processes. These proteins are able to recognize even suptable differences in the carbohydrate structure. Since they do not exhibit enzymatic activity, the binding partner remains unchanged. All these properties make lectins interesting objects for biomedical and biotechnological research. Although lectins in mushrooms occur with the same abundance as in other organisms, their research has received attention just recently. The reason is probably the recently discovered antiproliferative and immunostimulatory potential of these lectins. This thesis deals with potential lectins from Agaricus bisporus. The first part deals with bioinformatic analysis of sequences of hypothetical lectins obtained by previous laboratory research. Also included is the prediction of the structure of the most promising proteins. The second part is focused on the laboratory search and isolation of lectins from Agaricus bisporus var. brunensis by affinity chromatography and verifying the presence of "ricin-B-like" hypothetical lectin in this variant of the mushroom.
