Způsob degradace hostitelské kostní tkáně motolicemi rodu Troglotrema a hlísticemi rodu Skrjabingylus

Degradation of host osseous tissue by trematodes from genus Troglotrema and nematodes from genus Skrjabingilus

Abstract:

V této bakalářské práci se věnuji zástupcům rodu Troglotrema (třída Trematoda - motolice), zejména druhu T. acutum a zástupcům rodu Skrjabingylus (kmen Nematoda - hlístice), zejména druhu S. petrowi. Účelem této práce je seznámení s těmito parazitickými organismy, s možnými patologickými procesy v kostní tkáni a navržení způsobů, jakými mohou tito helminti degradovat kostní tkáň svých definitivních hostitelů (masožravé šelmy čeledi Mustelidae-lasicovití). U zástupců rodu Troglotrema a rodu Skrjabingylus není o aktivních účincích exkrečně-sekrečních produktů (ES produkty) prozatím nic známo. Předpokládá se však, že peptidázy, které jsou v ES produktech přítomny, hrají významnou roli při degradaci kostní tkáně v kraniální oblasti definitivních hostitelů motolic rodu Troglotrema a hlístic rodu Skrjabingylus. Testováním vzorků pomocí fluorimetrie a zymografie se podařilo naměřit aktivitu cysteinových peptidáz ve vzorku T. acutum. Ve vzorku S. petrowi byla prokázána minimální aktivita cysteinových peptidáz pouze pomocí fluorimetrie.

Abstract:

This thesis is focused on members of genus Troglotrema (class Trematoda), especially species T. acutum and members of genus Skrjabingylus (phylum Nematoda), especially species S. petrowi. The purpose of this thesis is to introduce these parasitic organisms in the context of possible pathological processes in the bone tissue and to suggest possibilities how could these helminths degrade bone tissue of their definitive hosts (carnivorous beast family Mustelidae-mustelids). Concerning the Troglotrema and Skrjabingylus genuses no functional protein molecules have been described and the composition of excretory-secretory products (ES products) is unknown. It is assumed, that peptidases which are possibly presented in the ES products could play an important role in the degradation of bone tissue in the cranial area of definitive hosts. We measured the peptidase aktivity in whole extracts of Troglotrema and Skrjabingylus samples by using fluorimetry and by zymography methods; presence of cysteine peptidase in T. acutum was recorded. The sample S. petrowi showed only minimal cysteine peptidase activity.
