Identification of microorganisms in food waste with a focus on culture methods

Abstract:

Plýtvání potravinami a s tím související nárůst produkce gastroodpadu je globální problém. Proto je nutné omezit plýtvání potravinami a pokusit se vzniklý gastroodpad zpracovat a znovu využít. Jedním z možných způsobů zpracování gastroodpadu je kompostování. Cílem kompostování je vytvořit kvalitní a bezpečný kompost, který může být následně použit jako přirozené hnojivo. Teoretická část práce je zaměřena na představení různých kompostovacích metod, charakterizaci fází kompostovacího procesu včetně změn bakteriální diverzity a hodnocení bezpečnosti a kvality kompostu. Dále jsou uvedeny také metody kultivace, detekce a identifikace bakterií. V praktické části práce byl zkoumán vliv aditiva Tea Tree oleje na množství a diverzitu bakterií během kompostovacího procesu s cílem posoudit jeho efektivitu při hygienizaci potravinového odpadu. Pro posouzení jeho účinku byla provedena kultivace s následnou izolací čistých bakteriálních kultur. Získané kultury byly podrobeny Gramovu barvení a KOH testu. Dále byl stanoven celkový počet KTJ/g vzorku na TSA agaru a na MRS Lactobacillus agaru. Z výsledků vyplývá, že Tea Tree olej snižuje množství bakterií a mění bakteriální diverzitu. Tyto výsledky tedy podporují hypotézu využití Tea Tree oleje pro hygienizaci potravinového odpadu.

Abstract:

Food wastage and the associated increase in the production of food waste is a global problem. It is therefore necessary to reduce food waste through processing and possible reuse. Composting is one of the possible ways of processing food waste. The composting process should result in a high-quality and safe compost that can be used as a natural fertilizer. The theoretical part of the thesis is focused on the introduction of different composting methods, the characterization of the phases of the composting process including changes in bacterial diversity, and the evaluation safety and quality of the compost. Methods for cultivation, detection and identification of bacteria are also presented. In the practical part of the thesis, the effect of the Tea Tree oil additive on bacterial abundance and diversity was investigated to assess its effectiveness in sanitizing food waste. To assess its effect, cultivation followed by isolation of pure bacterial cultures was carried out. The obtained cultures were subjected to Gram staining and KOH test. Furthermore, the total number of CFU/g of the sample was counted on TSA agar and MRS Lactobacillus agar. The results show that Tea Tree oil reduces the abundance of bacteria and alters the bacterial diversity. These results support the hypothesis of using Tea Tree oil for food waste sanitation.
