Symbiotic microorganisms of digestive tract of bryophagous insects

Abstract:

Endosymbionti v trávicím traktu umožňují zpřístupnit svému hostiteli živiny a výrazně ovlivňují komplexní biologické charakteristiky druhu. V této práci je ověřována využitelnost kultivační metody při zjišťování symbiotických mikroorganismů u bryofágů. Tím poukazuje na některá obecná úskalí kultivačních metod při zjišťování symbiotických mikroorganismů u hmyzu. U brouků z čeledi Byrrhidae byli na PDA agaru z trávicího traktu a zadečku izolováni endosymbionti. Rovněž byly izolovány mikroorganismy žijící na povrchu brouků. Mikroorganismy byly následně přiřazené k 34 morfotypům. Největší abundance a diverzita mikroorganismů byla dle očekávání zjištěna na povrchu těla brouků. V trávicím traktu a zadečku byla diverzita mikroorganismů stejná, avšak v trávicím traktu byla abundance více než šestkrát větší oproti zadečku. Součástí práce je rovněž návrh dalšího výzkumu s využitím alternativní metodiky.

Abstract:

The gut microbiota of insects facilitace the uptake of nutrients and significantly affect comprehensive biological characteristic of the host species. This thesis is a verification of the applicability for cultivation method to isolate symbiotic microorganisms of the bryophagous insects. It shows some of the general difficulties of the cultivation methods. Endosymbiont from a digestive tract and an abdomen from beatles of the family Byrrhidae were isolated on PDA agar. There were microorganisms living on the surface of the beetles isolated as well. Afterward were microoragnisms assigned to 34 morfospieces. The biggest abundance and diversity of the microoragnisms was not surprisingly discovered on the body surface of beetles. In the digestive tract and inside abdomen was a diversity of the microoragnism identical, however in the digestive tract the abundance was more than six time bigger compared to the abdomen. A part of this thesis is likewise a suggestion of additional research by using an alternative methodology.
