Laboratory isolation and cultivation of the zoonotic microorganism Borrelia burgdorferi sensu lato

Abstract:

Tato bakalářská práce se věnuje izolaci a kultivaci spirochet Borrelia burgdorferi sensu lato. Tyto gramnegativní mikroaerofilní bakterie se v poslední době vyskytují se stále větší prevalencí a mohou vyvolat závažné multisystémové onemocnění zvané Lymeská borrelióza. Bbsl může při infekci hostitele způsobit chronickou artritidu, kardiální problémy a zasahuje dokonce i centrální nervovou soustavu. Proto je také výzkum tohoto patogenu nezbytný. Nedílnou součástí výzkumu bakterií je správná izolace a kultivace. Za účelem správného pochopení těchto metod byly v práci shrnuty vybrané dosavadní znalosti o tomto procesu. Dále se tato práce zaměřuje na taxonomické zařazení Bbsl do systému bakterií, morfologii samotné spirochety, její životní cyklus a mechanismy perzistence v hostiteli. Také popisuje hlavní antigenní struktury těchto spirochet spojené s virulencí či jejím životním cyklem. V neposlední řadě jsou zde popsány symptomy LB a výzkum antimikrobiálních látek v rámci léčby.

Abstract:

This thesis deals with the isolation and cultivation of spirochetes Borrelia burgdorferi sensu lato. These Gram-negative microaerophilic bacteria have been increasingly prevalent in recent years and may cause a severe multisystemic disease called Lyme disease. Bbsl can cause chronic arthritis, cardiac problems, and even affect the central nervous system. Therefore, research on this pathogen is essential. Proper isolation and cultivation is an integral part of bacterial research. In order to understand these methods correctly, the present knowledge of the process has been summarized. Furthermore, this work focuses on the taxonomic classification of Bbsl into the bacterial system, morphology of the spirochete itself, its life cycle and mechanisms of persistence in the host. It also describes the major antigenic structures of these spirochetes associated with virulence or its life cycle. Last but not least, the symptoms of LD and the research of antimicrobials in the treatment are described.
