Proteomic and metabolomic approaches in plant biotic interaction analyses

Abstract:

Rostliny si jako přisedle žijící organismy musely vyvinout mechanismy, které jim pomáhají vyrovnat se s biotickými podněty. Tyto mechanismy zahrnují komplexy aktivních a pasivních obranných strategií vůči patogenům i procesy, které usnadňují interakci s prospěšnými organismy. Teoretická část bakalářské práce shrnuje biotické interakce rostlin a nejdůležitější metabolity podílející se na obraně rostlin. Experimentální část se skládá ze dvou nezávislých experimentů. V prvním experimentu byly analyzovány metabolomy topolů infikovaných patogenem rodu Phytophthora. Necílené profilování metabolomu identifikovalo několik domnělých markerů infekce, včetně katechinu a kyseliny skořicové. V druhém experimentu byla využita integrativní metabolomika a proteomika v analýze listu. Bylo zjištěno, že přibližně jedna třetina všech identifikovaných metabolitů a proteinů korelovala se stádiem vývoje listů.

Abstract:

Plants are sessile organisms, and as such, they had to develop mechanisms to cope with biotic stimuli. These mechanisms consist both of a complex mixture of active and passive defense strategies against pathogens, and of processes that facilitate interaction with beneficial organisms. The theoretical part of this thesis reviews plant's biotic interactions and summarizes the most important metabolites involved in plant defense. The experimental part consists of two independent experiments. The first experiment analyzed the metabolome of Phytophthora-infected poplar trees. An untargeted metabolome profiling identified several putative infection markers, including catechine and cinnamic acid. The second experiment employed integrative metabolomics and proteomics in an analysis of leaf. It was revealed that abundances of approximately one-third of all identified metabolites and proteins were correlated with the leaf developmental stage.
