Endocrine disruptive potential associated with human exposure to complex chemical mixtures

Abstract:

V této diplomové práci se zabýváme problematikou endokrinních disruptorů vyskytujících se ve vnějším a vnitřním ovzduší. Zaměřujeme se především na endokrinní disrupci signálních drah steroidních hormonů a kompletní dráhy thyroidních hormonů včetně jejich regulace syntézy, sekrece, metabolismu a transportu krví do místa působení. Při hodnocení negativních účinků endokrinních disruptorů jsme využili koncept tzv. „drah škodlivých účinků“ neboli Adverse Outcome Pathway (AOP), který propojuje účinky endokrinních disruptorů pozorované na molekulární úrovni in vitro s negativními účinky in vivo. V praktické části jsme provedli testování rekonstruovaných směsí vzorků prachových částic a plynné frakce ovzduší. Původní environmentální vzorky byly odebrány v městské lokalitě v zimním a letním období a ve venkovské lokalitě v zimním období. Takto odebrané vzorky byly frakcionací rozděleny na polární, semi-polární a nepolární frakci a byly testovány na estrogenní, androgenní a AhR-zprostředkovanou aktivitu. Cílem praktické části pak bylo porovnat aktivity původních environmentálních vzorků a námi testovaných rekonstruovaných směsí, které byly vytvořeny na základě chemických analýz původních vzorků prachových částic a plynné frakce. Rekonstruované směsi vždy byly tvořeny celkovou směsí všech stanovených polycyklických aromatických uhlovodíků a dále třemi frakcemi obsahující kyslíkaté, dusíkaté a parentální polycyklické aromatické uhlovodíky. Srovnáním biologických aktivit rekonstruovaných směsí s aktivitami původních environmentálních vzorků bylo zjištěno, že velkou část efektů především polární frakce prachových částic původních vzorků nebylo možné rekonstruovanými směsmi vysvětlit, a tudíž nejsou známy látky, které v těchto frakcích byly za pozorované efekty zodpovědné. Přepokládali jsme, že v polární frakci ovzduší se nacházejí především kyslíkaté deriváty polycyklických aromatických uhlovodíků, a proto jsme provedli testování jejich vybraných zástupů, jejichž přítomnost v ovzduší byla potvrzena. Zjistili jsme však, že námi testované kyslíkaté deriváty polycyklických aromatických uhlovodíků efekty polární frakce ovzduší z velké části nevysvětlují a pro identifikaci látek zodpovědných za pozorované efekty je potřeba dalšího výzkumu. V další části praktické části bylo provedeno testování vybraných látek relevantních pro každodenní lidskou expozici pro jejich schopnost aktivace thyroidního receptoru. Nejvýznamnější efekty byly pozorovány u látek BDE47, BDE99, BP2, BPA, TBBPA a TCS. U ostatních testovaných látek nebyla aktivace TR pozorována. Je však zřejmé, že řada látek vyskytujících se ve vnitřním prostředí může v endokrinní disrupci thyroidního systému hrát roli.

Abstract:

In this thesis we deal with the issue of endocrine disruptors occurring in outdoor and indoor air. We focus primarily on endocrine disruption of steroid hormone signal pathways and complete thyroid hormone pathways, including their regulation of synthesis, secretion, metabolism and transport to the site of action. In evaluating the negative effects of endocrine disruptors, we used the concept of the so-called Adverse Outcome Pathway (AOP), which links the effects of endocrine disruptors observed at the molecular level in vitro with the negative effects in vivo. In the practical part, we performer testing of reconstructed mixtures of dust particles and gaseous air fraction of ambient air. The original environmental samples were taken at the urban site in winter and summer and at the rural site in winter. The samples were fractionated into polar, semi-polar and non-polar fractions and tested for estrogenic, androgenic and AhR-mediated activity. The aim of the practical part was to compare the biological activities of the original environmental samples and the reconstructed mixtures, which were created on the basis of chemical analyses of the original samples of dust particles and gaseous fraction. The reconstructed mixtures always consisted of a total mixture of all determined polycyclic aromatic hydrocarbons and three fractions containing oxygen, nitrogen and parent polycyclic aromatic hydrocarbons. By comparing the biological activities of the reconstructed mixtures with the activities of the original environmental samples, it was found that a large part of the effects, especially the polar fraction of dust particles of the original samples could not be explained by the reconstructed mixtures. We assumed that the polar fraction of the air contains mainly oxygen derivatives of polycyclic aromatic hydrocarbons, and therefore we performed testing of their selected representatives, whose presence in the air was confirmed. However, we found that the oxygen derivatives of polycyclic aromatic hydrocarbons tested by us do not largely explain the effects of the polar fraction of the air, and further research is needed to identify the substances responsible for the observed effects. In the next part of practical part, testing of selected substances relevant to everyday human exposure for their ability to activate the thyroid receptor was performed. The most significant effects were observed for BDE47, BDE99, BP2, TBBPA and TCS. TR activation was not observed for the other tested substances. However, it is clear, that many substances present in the indoor environment may play a role in the endocrine disruption of the thyroid system.
