The effect of forskolin derivatives on human neutrophil activity

Abstract:

Předkládaná diplomová práce se zabývá vlivem forskolinu a jeho derivátu colforsinu na aktivitu lidských neutrofilů, které mají rozhodující roli v obraně organismu proti cizorodým látkám, jakož i při lokálním či systematickém zánětu. Forskolin je diterpen izolovaný z indické rostliny Coleus forskolhii, který má unikátní vlastnost rychle a přímo aktivovat adenylátcyklasy (AC). Ty jsou schopny syntetizovat významného sekundárního posla - cyklický 3´,5´-adenosinmonofosfát (cAMP), jehož hlavní funkcí je regulovat množství intracelulárních procesů, mezi které lze zařadit inhibici apoptózy a produkci reaktivních metabolitů kyslíku (RMK) lidských neutrofilů. Cílem práce bylo objasnit vliv forskolinu a jeho derivátu colforsinu na aktivitu lidských neutrofilů vyizolovaných z krve a následně porovnat účinek těchto dvou molekul navzájem. Jako aktivátor neutrofilů byl použit N-formyl-methionyl-leucyl-fenylalanin (fMLP, 1×10-6 M). Kromě testovaných látek aktivujících AC byl sledován také vliv 3-isobutyl-1-methylxanthinu (IBMX, 5x10-5 M), který zvyšuje cAMP inhibicí fosfodiesteras. Z výsledků diplomové práce vyplývá, že forskolin a jeho derivát colforsin (5x10-5 M) jsou schopny zvýšením intracelulární koncentrace cAMP inhibovat aktivitu neutrofilů, konkrétně produkci RMK (chemiluminiscence) a degranulaci laktoferrinu (ELISA). Dále bylo zjištěno, že inhibice produkce RMK nebyla způsobena antioxidačními vlastnostmi testovaných aktivátorů AC. Při vzájemném porovnání efektů se jako účinnější inhibitor aktivity neutrofilů jevil colforsin. V kombinaci s IBMX byl inhibiční účinek obou látek prohlouben. Pozorované účinky forskolinu ani colforsinu nebyly dány změnou viability buněk. Závěrem lze říci, že použitím selektivních aktivátorů AC lze docílit inhibice neutrofilních funkcí. Tohoto efektu je možné využít v patologických stavech, ve kterých neutrofily přispívají k poškození vlastní tkáně.

Abstract:

Presented diploma thesis deals with the influence of forskolin and its derivative colforsin on the activity of human neutrophils, which have a decisive role in defending the organism against heterogeneous pathogens, as well as in local or systematic inflammation. Forskolin is a diterpen isolated from the Indian plant Coleus forskolhii, which quickly and directly activates adenylcyclases (AC). ACs are able to synthesize an important secondary messenger cyclic-3´,5´-adenosinemonophosphate (cAMP), which main function is to regulate various intracellular processes including inhibition of apoptosis, and the production of reactive oxygen species (ROS) of human neutrophils. The aim of this study was to clarify the effect of forskolin and its derivative colforsin on the activity of human neutrophil isolated from blood and then compare their effects mutually. N-formyl-methionyl-leucyl-phenylalanine (fMLP, 1×10-6 M) was used as an activator of neutrophils. Besides the tested activators of AC we investigated also the effect of 3-isobutyl-1-methylxanthine (IBMX, 5x10-5 M) which increases cAMP by inhibition of phosphodiesterases. The results showed that forskolin and its derivative colforsin (5x10-5 M) were able to inhibit the activity of neutrophils, particularly ROS production (chemiluminescence) and degranulation of lactoferrin (ELISA) by increasing the intracellular concentration of cAMP. Inhibition of ROS production was not due to antioxidant activities of tested AC activators. Interestingly, colforsin was more effective than forskolin. Combination of both tested compounds with IBMX showed advanced inhibitory effect. Neither forskolin nor colforsin decreased the viability of neutrophils. In conclusion, we showed that selective activators of AC are able to inhibit neutrophil functions. This may be beneficial in pathologies where neutrophils contribute to tissue injury of the host.
