Abstract:

Ciel'om tejto diplomovej práce je urcenie optických vlastností, presnejšie optických konštánt amorfnej fázy disulfidu molybdénu, a zároven rozšírenie a posúdenie nadobudnutých hodnôt a výsledkov optických konštánt med'natého ftalocyanínu, ktorým sme sa zaoberali v bakalárskej práci. K urceniu optických konštánt bola využitá v oboch prípadoch metóda spektroskopickej elipsometrie založenej na zmene polarizácie svetla po odraze na rozhraní dvoch rôznych médií. Disulfid molybdénu vo fototermálnej terapii nesie rolu tzv. fototermálneho transdukcného činidla, čím je látka absorbujúca svetlo v určitých vlnových dlžkach, ktoré premiena na teplo, pomocou ktorého dochádza k ablácii nádorových buniek. Naproti tomu s ftalocyanínom sa stretávame vo fotodynamickej terapii, kde nesie úlohu fotosenzitizéra, teda látky absorbujúcej energiu svetla, ktorú prenáša okolitému kyslíku v tkanive, kde dochádza k vytvoreniu jeho reaktívnych foriem -ROS, ktoré usmrcujú nádorové bunky. Zamerali sme sa na oblasti terapeutického (biologického) okna každej z látok, teda oblasti maximálnej penetracnej hlbky v danom tkanive. U ftalocyanínu a využíva spektrálna oblast' UV/VIS (620-800 nm), kým u disulfidu je to NIR oblast' rôznych vlnových dlžok. Experimentálne urcené optické konštanty boli parametrizované adekvátnou matematickou funkciou a d'alej porovnané s hodnotami prezentovanými v literatúre. Toto porovnanie dáva možnost' k vyladeniu ci už teoretických (ab-initio) ale aj praktických postupov, a tým zlepšit' celkovú protinádorovú terapiu ako celok.

Abstract:

The aim of this thesis is to determine the optical properties, more precisely the optical constants of the amorphous phase of molybdenum disulfide, and to extend and evaluate the acquired values and results of the copper phthalocyanin optical constants that we have dealt with in the baccalaureate work. In both cases, the spectroscopic elliptometry method based on the variation of light polarisation after reflection at the interfaces of two different media was used to determine the optical constants. The disulfide of molybdenum in photothermal therapy is a so-called photothermal transducer, which is a light-absorbing substance in certain wavelengths that converts to heat to abate tumour cells. By contrast, phthalocyanine is found in photodynamic therapy, where it acts as a photosensitiser, a substance that absorbs the energy of light, which transmits it to the surrounding oxygen in the tissue, where it forms its reactive form - ROS, which kills cancer cells. We focused on the areas of the therapeutic (biological) window of each substance, the areas of maximum penetration depth in the tissue. For phthalocyanine and uses the spectral UV / VIS range (620-800 nm), while for disulfide it is an NIR range of different wavelengths. The experimentally determined optical constants were parameterized by an adequate mathematical function and further compared with the values presented in the literature. This comparison provides an opportunity to fine-tune both theoretical (ab-initio) and practical practices, thereby improving overall anticancer therapy as a whole.
