Abstract:

Využití mtDNA pro charakterizaci populačních struktur druhů rodů Gobio a Romanogobio Taxonomie a systematika druhů rodů Gobio a Romanogobio je v současné době velmi nepřehledná. O validitě řady druhů, poddruhů a forem, jež byly popsány v minulosti, se vedou diskuse. Současná systematika obou rodů je založena především na morfologických charakteristikách a nebyla dosud potvrzena molekulárními analýzami. Tato studie se zabývá fylogenetickými vztahy 28 druhů obou rodů v euroasijském kontextu. Genetická diverzita hrouzků byla zkoumána technikami RAPD a přímým sekvencováním control regionu, cytochromu b a intronu S7 r-proteinu. Molekulární analýzy potvrdily rodovou diferenciaci původního rodu Gobio na dva rody Gobio a Romanogobio. Bylo prokázáno, že území ČR osídlují druhy Gobio gobio, Gobio obtusirostris, Gobio carpathicus (ojediněle), Romanogobio banaticus, Romanogobio vladykovi, Romanogobio belingi a druh označený termínem „species-in-waiting“ Romanogobio carpathorossicus. Na území SR se vyskytuje vedle tří výše jmenovaných druhů rodu Gobio i čtvrtý druh pracovně označený Gobio sp. 1. Rod Romanogobio má obdobnou druhovou skladbu jako v ČR vyjma druhu Romanogobio belingi. Na slovenském území byl navíc potvrzen výskyt druhu Romanogobio friči. Výsledky ukázaly, že řeky Černého moře vykazují větší druhovou pestrost rodu Gobio než řeky tekoucí do Severního a Baltského moře. Celkem bylo zachyceno 16 hybridních událostí, které se týkaly obou rodů. Na území SR byla vymezena hybridní zóna v oblasti severních přítoků řeky Tisy. Byla posouzena genetická odlišnost populací na území ČR a SR, především u kriticky ohrožených druhů rodu Romanogobio. U šesti populací druhu Gobio gobio a dvou populací Gobio obtusirostris byla zjištěna statisticky významná mezipopulační diverzita. Bylo odhaleno celkem 40 populačně diagnostických RAPD markerů. Byla nalezena jednoduchá rozlišovací metoda nazvaná „S7indel diagnostika“, která se osvědčila jako nástroj rychlé identifikace druhů rodu Gobio. Bylo navrženo rozšíření subjektu ochrany o „ochranu genetické diverzity“ jako v současnosti nejsnáze uplatnitelný postup při ochraně biodiverzity v ČR.

Abstract:

The using of mtDNA for characteristics of population structures of species from the genera Gobio and Romanogobio Taxonomy and systematics of species of the genera Gobio and Romanogobio is rather confused at present. Validity of a number of species, subspecies and forms described in the past is under discussion. The present systematics of both the genera is based especially on morphological characteristics and has not been confirmed by molecular analyses yet. The study deals with phylogenetic relationships of 28 species of both the genera in the Euro-Asian context. Genetic diversity of gudgeons was investigated using the RAPD technique and direct sequencing of the control region, cytochrome b and the intron of the S7 r-protein. Molecular analyses have confirmed generic differentiation of the original genus Gobio into two genera Gobio and Romanogobio. It has been demonstrated that the territory of the Czech Republic is inhabited by the species Gobio gobio, Gobio obtusirostris, Gobio carpathicus (only one specimen found), Romanogobio banaticus, Romanogobio vladykovi, Romanogobio belingi and by the species Romanogobio carpathorossicus which is denoted as a “species-in-waiting”. Concerning the territory of the Slovak Republic, in addition to the three above mentioned species of the genus Gobio also a fourth one occurs there which is provisionally called Gobio sp. 1. The genus Romanogobio has a similar species composition as in CR except for Romanogobio belingi. In addition, the presence of Romanogobio friči was confirmed in the territory of Slovakia. The results have pointed out, that the rivers of the Black Sea show greater species diversity of the genus Gobio than the rivers flowing into the North and Baltic Seas. Altogether 16 hybridization events concerning both the genera were recorded. In the territory of the Slovak Republic, a hybrid zone was delineated in the area of nothern tributaries of the Tisa River. Genetic difference of the populations in the territories of CR and SR was examined, especially in the critically endangered species of the genus Romanogobio. Statistically significant inter-population diversity was found in six populations of Gobio gobio and two populations of Gobio obtusirostris. Altogether 40 population-diagnostic RAPD markes were identified. A simple differentiation method called “S7indel diagnostics” was found; it proved to be a useful tool for quick identification of the species of the genus Gobio. Broadening of the subject of conservation with “genetic diversity conservation” was suggested as the easiest procedure in biodiversity conservation in CR at present.
